Objective:To explore the changes of gastric mucosa status after lianpu Yin modified decoction intervention in chronic atrophic gastritis rat model,and to explore the correlation between lianpu Yin Plus flavor Prescription intervention in chronic atrophic gastritis rat model and intestinal flora.Methods:80 healthy SPF SD rats were selected,and 5 rats were randomly selected as the blank control group(group A).The other 75 rats were used for modeling: free drinking of 100 mg/ L MNNG solution,intragastric administration of 0.03 g / kg ranitidine solution every day,abnormal hunger and satiety,eating for 2 days,fasting for 1 day,intragastric administration of 56 ℃ 150 g / L Na Cl solution in the morning of the first day,intragastric administration of 2ml every 100 g,intragastric administration of 30% ethanol in the afternoon of the day of fasting,and intragastric administration in the 6th,8th,10 th weeks At 12 and 14 weeks,two model rats were randomly selected.The gastric tissue was stained with hematoxylin eosin(he).The pathological changes of gastric tissue were observed under light microscope.After confirming the success of the model,the rat model of chronic atrophic gastritis was obtained.Sixty model rats were randomly divided into model group(group B),western medicine group(vitamin group)(Group C),lianpuyin original formula group(Group D),modified decoction low-dose group(Group E),modified decoction medium-dose group(Group F)and modified decoction high-dose group(group G),with 10 rats in each group.After the successful replication of the model,the intervention of traditional Chinese medicine was given by gavage on the second day,once a day for 8 weeks.The dose of liquid medicine is converted into the dose of experimental rats and adults(60 kg),1 ml / 100 g,and the gavage dose is 2.084 g / 100 g / D respectively.The blank control group and model group are perfused with equal doses of normal saline.The rats fasted water for 24 hours after the last administration,and were anesthetized by intraperitoneal injection of 10% chloral hydrate solution.The rats were killed,and the fecal liquid nitrogen was taken for preservation and examination.Rat feces were detected and analyzed based on 16 S r DNA high-throughput sequencing technology.Results:In group A,the gastric mucosal cells were arranged orderly,the proper glands were arranged orderly,and the shape was regular.There was no reduction of glands,intestinal metaplasia and intraepithelial neoplasia;In group B,the epithelium of gastric mucosa shrank and thinned,the number of glands decreased,a large number of inflammatory cell infiltration,congestion and edema and intestinal epithelization were observed.In group C,the degree of inflammatory cell infiltration decreased,glands atrophied,and intestinal metaplasia was still accompanied.D.The number of glands in Group E was increased.F.In group G,the degree of inflammatory infiltration decreased significantly,the number of glands increased and arranged neatly.The result of group F was significantly better than that of group G.The intestinal flora of normal rats mainly includes Firmicutes,Bacteroidetes,superphylum,Proteus and verruca.The dominant bacteria families are spirillaceae,prevotellaceae,Treponema,rumen coccaceae and lactobacilli.After atrophic lesions of gastric mucosa,at the gate level,the proportion of verrucous microflora increased,and the proportion of thick walled bacteria and super gate decreased;At the family level,the abundance of spirillaceae,prevotellaceae,Treponema and rumen coccaceae in group A was significantly lower than that in group A.After the drug intervention,the levels of spirillaceae and treponceae in each group were higher than those in group B,and the abundance proportion of the top ten species in groups e,F and G gradually increased,close to that in group A.It preliminarily shows that lianpuyin modified formula can play a therapeutic role by regulating the structure of intestinal flora.At the same time,this study predicted the function of intestinal flora.The results showed that the intestinal flora of CAG rat model was weaker than that of normal rats in terpene and polypeptide metabolism,energy metabolism,antimicrobial resistance,cell activity,carbohydrate metabolism and amino acid metabolism.After drug intervention,the metabolism of terpene and polypeptide,energy metabolism,antimicrobial resistance,carbohydrate metabolism Amino acid metabolism was enhanced in different degrees compared with group B.in terms of carbohydrate metabolism,group F had the best recovery level;In amino acid metabolism,group D,e,F and g were better than group B and C;In the ways of exogenous biodegradation and metabolism,metabolism of other amino acids,metabolism of cofactors and vitamins,biosynthesis and metabolism of glycans,the flora of group B was stronger than that of group A;After drug intervention,the metabolism of other amino acids in groups D,e,F and G decreased,and group F performed best;The biosynthesis and metabolism of cofactors,vitamins and glycans in group D,e and F were lower than those in group B,and those in group G were higher than those in group B.It preliminarily shows that lianpuyin modified formula has a certain correlation between intervening in CAG rat model and improving the function of intestinal flora.Conclusion:(1)Lianpuyin modified decoction has definite curative effect in blocking the transformation of CAG inflammation and cancer,reducing the inflammatory environment of gastric mucosa and repairing gastric mucosa.(2)The mechanism of lianpuyin modified decoction in CAG rat model is highly correlated with the improvement of intestinal flora.
