Phellodendron amurense Rupr.is a traditional Chinese medicinal plant.It is used as medicine with its dry bark,which has important medicinal value.It is a representative medicine for clearing heat and drying dampness.It also has antioxidant activity,anti-inflammatory and analgesic effects,anti gout and other pharmacological effects.However,the research on other parts of P.amurense is still blank.In order to protect the resources of P.amurense and make sustainable development and utilization of the medicinal value of it,the anti gouty arthritis activity and auxiliary pharmacological action of P.amurense leaves and its different processed products were studied on the basis of consulting a large number of literatures.The details were as follows:Review part: the herbal records,resource distribution and protection,chemical composition,pharmacological action,development and utilization of P.amurense,as well as the research progress of anti gouty arthritis drugs were reviewed.The research part were as follows:1.The preparation technology of black tea and green tea made by P.amurense leaves was studied.2.The contents of total alkaloids and flavonoids in P.amurense leaves and its different processed products were determined by colorimetry,which was compared with P.amurense barks.The results showed that the highest content of alkaloids was P.amurense barks,and the highest content of total flavonoids was fermented fried P.amurense leaves3.Anti gouty arthritis activity in mice: the acute gouty arthritis(AGA)model was established by injecting sodium urate(MSU)into the ankle joint of mice.The perimeter of ankle joint was measured,the pathological morphology of kidney and synovium was observed by HE staining,the expressions of NALP3,caspase-1 and ASC in synovial tissue were detected by Western blot,and the m RNA expression of IL-1β and TNF-α in synovium was detected by real time PCR.The results showed that the anti gouty arthritis effect of fermented fried P.amurense leaves was the best,and there was no significant difference with the positive group.It can significantly reduce joint swelling,improve renal sodium urate crystal and synovial tissue inflammatory infiltration,reduce UA in serum,IL-1β and TNF-α in synovial membrane and XOD in liver,and reduce IL-1β and TNF-α m RNA expression in synovial membrane.Fried P.amurense leaves,original P.amurense leaves and P.amurense bark also have different degrees of anti acute toxicity gouty arthritis,and showed a good dose relationship.4.Adjunctive pharmacological experiments(1)Antioxidant activity research: the antioxidant activity of P.amurense leaves and its different processed products was evaluated by the scavenging rate of DPPH free radical,ABTS free radical,hydroxyl free radical,superoxide anion free radical and the determination of iron atom reducing power.The results showed that the fermented fried P.amurense leaves had the best antioxidant activity,the scavenging rates of DPPH free radical,ABTS free radical,hydroxyl free radical and superoxide anion free radical,and the reducing ability of iron atom were higher than those of the other three samples,which were the closest to vitamin C.The results showed that the antioxidant activity of the samples was positively correlated with the content of total flavonoids,and flavonoids had good antioxidant activity,which accorded to the literature.(2)Anti inflammatory and analgesic activities: egg white induced paw swelling,xylene induced auricle swelling,hot plate induced pain and acetic acid writhing were used to study the anti-inflammatory and analgesic activities of P.amurense leaves and its different processed products.The results showed that the fermented fried P.amurense leaves had the best anti-inflammatory and analgesic activities,which had little difference with the positive group,and could significantly reduce the inflammatory factor malondialdehyde(MDA)in serum of mice.The results showed that the contents of PGE2,IL-1 and SOD in serum of mice were increased,which corresponded to the best antioxidant activity of the fermented fried P.amurense leaves.(3)Acute toxicity test: the acute toxicity test of P.amurense leaves and its different processed water extracts was conducted for the first time.The results of pre-test showed that there was no death and LD50 in each sample group after 7 days of administration,so the maximum tolerated dose method was used for acute toxicity test,and no death was found in the mice after 14 days of continuous observation.The mice were in good condition,and there was no abnormal change in body weight and organ index.In conclusion,it was found that the original P.amurense leaves and its different processed products,namely fermented fried P.amurense leaves,had good antioxidant activity,anti-inflammatory and analgesic activities and anti acute gouty arthritis activities.The overall pharmacological activity was better than that of P.amurense bark.The results of acute toxicity test showed that the P.amurense leaves and its processed products had no acute toxicity and could be reasonably developed and utilized.This can not only protect the wild P.amurense tree resources,but also provide a theoretical basis for the research of new anti gouty arthritis drugs.At the same time,the product development of black tea and green tea made by P.amurense leaves is also a great significance to the characteristic health industry in Northeast China.
