Phellodendron amurense is listed as top grade in "Shen Nong’s Materia Medica".At present,in the 2015 edition of "Chinese Pharmacopoeia",only two components of Phellodendron amurense,namely berberine and phellodendron,off Phellodendron amurense and palmatine hydrochloride The content standard cannot control its quality comprehensively.In this paper,the preparation process of Phellodendron amurense extract is optimized,and the fingerprint is established by UPLC method.The 8 components(berberine,phellodendrine,magnolia base,berberine,jatrorrhizine hydrochloride,Berberine,palmatine hydrochloride,chlorogenic acid)were analyzed quantitatively,which provided a reference basis for comprehensive control of the quality of Phellodendron chinense and extracts.Through static adsorption method,the resin for purification of Phellodendron chinense is ADS-7 macroporous resin.The amount of sample required for the test medicinal materials,the water for elution,and the concentration and amount of alcohol used during elution are investigated by the dynamic adsorption test.The orthogonal experiment method was used to investigate the concentration of the drug solution,the sample loading rate,and the inner diameter of the chromatographic column.Finally,the purification process of Phellodendron chinense was determined as follows: the ratio of the amount of medicinal materials and the amount of resin was 1: 3,and the loading concentration and the inner diameter of the chromatographic column were not limited.5 BV purified water for elution,9 BV90% ethanol as eluent.In this paper,a UHPLC method was established to quickly analyze the content of 8 active ingredients in Phellodendron chinense in 10 minutes.The methodological verification shows that the average sample recovery rate(Average sample recovery rate)and relative standard deviation(RSD)of Phellodendron chinense and Phellodendri chinensis: 99.35% ~ 100.30%,0.12% ~ 0.72%;99.74% ~100.42%,0.80% ~ 2.27%.This method is fast,stable and reliable.Using this method,the content of 30 batches of Phellodendron amurense and 5 batches of Phellodendron amurense was determined,and the content of the medicinal materials in each place of origin and batch was very different.It is recommended to add ingredients such as magnolia base,berberamine,and jatrorrhizine hydrochloride on the basis of the original content determination standard of the 2015 edition of "Chinese Pharmacopoeia"(otherwise,berberine produced by Phellodendron chinense should also be added as the standard for processed products,The content of berberine is not less than 0.03%.)The content of berberamine,magnolia and jatrorrhizine in Sichuan Phellodendron chinense(quality standard)is not less than0.10%,0.08% and 0.02%;,Magnolia base,and jatrorrhizine content were not less than 0.03%,0.02%,0.035%.Using the "Chinese Medicine Chromatography Fingerprint Similarity Evaluation System(2012 Version)" software to analyze the similarity of the data,the Chuanhuangbai(the 14 th produced in Shizhu County,Chongqing)map was used as a reference to generate a fingerprint of Chuanhuangbai medicine;33)Guan Huangbai(Liaoning)analysis map is used as a reference map togenerate fingerprint map of Guanhuangbai medicine.The similarity of the collected batches of medicinal materials is greater than or equal to 0.994,and the similarity is better.It can be used to control the quality of Phellodendron chinense and its extracts. |
