Objective: Glioma characterized by high degree of disability,is an extremely aggressive and devastating carcinomas in the brain,which playing an important role in the NeuroOncology rehabilitation.Based on the fundamental research,our research is aimed to provide new ideas for improving the prognosis of glioma patients and promoting the development of Neuro-Oncology rehabilitation via exploring the pathogenesis of glioma and searching for new therapeutic strategies.Centrosome amplification together with Warburg effect have been verified as malignant biological characteristics of tumors.Phosphoglycerate mutase 1(PGAM1)regulates the key reaction of the Warburg effect.Studies have showed that PGAM1 is associated with the proliferation and metastasis in various tumors and promote the disease progression.At present,a few studies about the relationship between them have taken.In a word,we will observe the expression of PGAM1 and γ-tubulin at the level of glioma paraffin specimen tissues,and preliminarily explore the relationship between PGAM1 and centrosome amplification in glioma,as well as the clinical significance.Methods: 78 patients with diagnosed Glioma after surgery were elected in the study in our hospital from 2016 to 2020.Meanwhile,we collected 10 non-tumor brain tissue specimens from patients with cerebral trauma or cerebral hemorrhage who underwent intracranial decompression.All tissues specimens were examined with HE staining.We will observe the protein expression levels of PGAM1,Ki-67 as well as γ-tubulin,and view centrosome amplification by means of immunohistochemistry and immunofluorescent staining respectively.Additionally,the correlation between PGAM1 and γ-tubulin,and their association of the clinicopathological features were analyzed by Spearman correlation analysis.Results: HE staining showed tumor cells display different features.The expression levels of PGAM1 and γ-tubulin as well as Ki67 in high grade glioma exceeded in these in low grade glioma significantly,and were obviously higher than those in normal brain tissues.(P< 0.05).Immunofluorescence analysis showed that centrosome amplification was also detected in glioma.As the pathological grade of the tumor go up,the rate of centrosome amplification also increases(P< 0.05).A positive correlation between PGAM1 and γ-tubulin protein expression on behalf of centrosome amplification in glioma tissues was discovered by Spearman correlation analysis(r = 0.621,P <0.05).Moreover,the expression of PGAM1 and γ-tubulin were closely related to the WHO grade of glioma and the expression level of Ki-67 recognized as the proliferation indice(P< 0.05),but there was no significant impact on other factors(P > 0.05).Conclusion: The expression levels of PGAM1 and γ-tubulin are increased significantly in glioma and closely related to the WHO pathological grade and Ki67.There is a positive correlation between the two proteins.The results show that PGAM1 and centrosome amplification are positively correlated,which may act an important part in glioma.
