Objective: To observe and compare the effects of different types of cardioplegia on the proliferation of immature cardiomyocytes in isolated neonatal mice.Methods: The hearts of two day-old newborn mice were isolated and cultured in vitro by means of differential velocity adherent method,Obtain high-purity cardiomyocytes after 48 h,Randomly divide the obtained neonatal mouse cardiomyocytes into 5 groups.Each group was given modified St.Thomas solution(STH),HTK solution,imitation del Nido solution and compound electrolyte solution for 2 hours and continued to cultivate for 24-72 hours.The proliferation of cardiomyocytes was observed by technique of EdU fluorescence staining,Ki-67 fluorescence staining and MTT colorimetry.The expression of YAP1 protein was detected by Western blot.Results: Ki-67 immunohistochemical staining and EdU fluorescence staining positive cell count results:The HTK group was the highest,followed by the modified STH group,and the del Nido analogue group was lower than the compound electrolyte group.OD value detected by MTT method: The HTK group was the highest,followed by the modified STH,and the del Nido analogue group was higher than the compound electrolyte group.YAP1 western blot gray value of HTK group was higher than other groups.Conclusion: HTK solution has a positive protective effect on the proliferation of neonatal mouse cardiomyocytes cultured in vitro.Modified STH solution is also an excellent crystal cardioplegia for immature myocardium.
