| Background:The innovative theory of Glucolipid Metabolic Disorders(GLMD)was initiated by Professor Guo Jiao.It is a complex disease characterized by the disorder of glucose and lipid metabolism,involving many factors such as heredity,environment and spirit.Shenqi Jiangtang Granule as a compound preparation of traditional Chinese medicine has the characteristics of “multi-component,multi-target and multi-channel”.In the previous research,Shenqi Jiangtang Granule was found to moderated glucose and lipid metabolism.However,its material basis and mechanism of action have not been studied in depth.The purpose of this study is to study the chemical basis,Network pharmacology and Short Chain Fatty acid targeted metabonomics of Shenqi Jiangtang Granule,and to explore the mechanism of its treatment of GLMD.Objective:The purpose of this study is to clarify the material basis of Shenqi Jiangtang granules and to elaborate its mechanism of action in the treatment of Glucolipid Metabolic Disorders.Methods:The fingerprint of Shenqi Jiangtang Granule was established.The non-volatile and volatile components in Shenqi Jiangtang Granule were identified by UPLC-Q-TOF-MS and GC-MS.Through Network pharmacology and Short Chain Fatty Acids targeted metabonomics system,the mechanism of Shenqi Jiangtang Granule in the treatment of GLMD was clarified.Results:1.Shenqi Jiangtang Granule material basis results(1)Study on fingerprint of Shenqi Jiangtang granuleThrough the establishment of the UPLC fingerprint of Shenqi Jiangtang Granule,a total of 44 common peaks were obtained,the similarity of 24 batches of Shenqi Jiangtang Granule were more than0.95,indicating that the quality of 24 batches of Shenqi Jiangtang Granule was consistent.(2)The results of qualitative study on the chemical components of Shenqi Jiangtang GranuleOne hundred and thirty-two,including saponins,flavonoids and lignans,were identified by ultra high performance liquid chromatography tandem quadrupole time of flight mass spectrometry(LC-MS).Eighty six volatile components,including hydrocarbons,alcohols,aldehydes,phenolic acids,lignans and fatty acids,etc.were identified by GC-MS.2.Effect of Shenqi Jiangtang Granule on Glucolipid Metabolic Disorders(1)Network pharmacology resultsThe results of network pharmacology and molecular docking showed that Ginsenoside Rh1,Ginsenoside RH4 and TNF,ACE;Acylalisol B,Pachymicacid and ACE,CETP;Calycosin-7-glucoside,Tillioside and ALB,PPARγ,LDLR,PPARα had good binding activity,which was the potential effective component and core target of Shenqi Jiangtang Granule in the treatment of GLMD.Shenqi Jiangtang Granule plays an important role in the treatment of GLMD by regulating neuroendocrine disorders,insulin resistance,inflammation and other related targets and pathways.(2)Short Chain Fatty Acids targeted metabonomics resultsEight kinds of SCFAs were detected in the faeces of rats.Compared with the normal group,acetic acid,propionic acid and butyric acid were significantly increased,isobutyric acid and isovaleric acid were significantly reduced in the model group,and there was no significant change in valeric acid,hexanoic acid and heptanic acid.After the treatment of Shenqi Jiangtang Granule in high doses,isovaleric acid was significantly increased.Conclusion:In this study,the fingerprints of Shenqi Jiangtang Granule were established,and qualitative analysis was carried out to provide the research basis for the further development and utilization of this variety.From the perspective of network pharmacology,the integration mode and mechanism of “multi-component,multi-target and multi-channel” of Shenqi Jiangtang Granule in the treatment of GLMD were preliminarily analyzed.Molecular docking showed that the binding activity of Ginsenoside RH4 and ACE,Calycosin-7-glucoside and PPARα,Tilloside and PPARγ was higher than that of positive drugs Benazepril and ACE,Lifibrate and PPARα,Rosiglitazo and PPARγ With high activity,it is expected to become a new drug for reducing blood pressure,lipid and glucose.Short Chain Fatty Acids targeted metabonomics study found that high dose Shenqi Jiangtang Granule can significantly increase the content of isopentanoic acid in the feces of T2 DM rats.In conclusion,this study suggests that Shenqi Jiangtang Granule may improve the metabolism of glycolipid and maintain the stability of intestinal environment by regulating insulin resistance,inflammation and the metabolism of SCFAs,especially isovaleric acid in the intestine,which provides a new idea and experimental basis for the mechanism of Shenqi Jiangtang Granules in the treatment of GLMD. |
