Abstract/Summary:
Objective:The model of extravasation skin injury was established by subcutaneous injection of amiodarone in SD rats.To investigate the effect of chrysin on skin injury induced by amiodarone extravasation in rats.The tissue around the extravasation area was collected at different time points,and the effect of chrysin on tissue injury caused by amiodarone extravasation was discussed from the aspects of histopathology and cytokine biology,and the basic medicine was integrated into the nursing work,Which will provide theory basis for clinical infusion extravasation nursing work.Method:1.Establishment of amiodarone extravasation skin model in SD rats.The rats were randomly assigned into the following groups: 5% GS control group,12.5mg/ml low concentration amiodarone group,25mg/ml medium concentration amiodarone group and 50mg/ml high concentration amiodarone group.The liquid extravasation models were established by subcutaneous injection of 0.5ml 5% GS,12.5mg/ml amiodarone,25mg/ml amiodarone and 50mg/ml amiodarone respectively.Through the dynamic observation of the skin changes of the injection site,the calculation of the injury area and the score of the injury degree,and the analysis of histopathological changes at different time points,we analyzed whether amiodarone extravasation damaged the skin or node.And select the appropriate concentration of injury model for the further research.By collecting the images and injury area on day 0,3,7 and 10,and collecting the tissue around the injection site of rats in each group.On day 0,3,7 the tissue of the lesion was collected for H&E staining.It was used for histopathological changes determinationl to observe changes to determine whether amiodarone extravasation will cause skin damage and choose the appropriate modeling concentration.2.The rat model of skin extravasation injury was established by subcutaneous injection of 0.5 ml of amiodarone.After successful modeling,these rats were randomly assigned into the following groups:control group,10%DMSO group,and low-dose,medium-dose and high-dose chrysin groups(10,20 and 40 mg/ml).The extravasation injury model was given locally on the same day for seven days.On day 0,3,7and 10 of administration,the image of the lesion was taken to calculate the area,and the tissue of the lesion was collected for H&E staining.Then,the content of IL-6 and TNF-α was measured by ELISA,and the protein expression level of b FGF in the wound tissue was detected by immunohistochemical staining.Results:1.After half an hour of injection,different degrees of skin injury occurred in all groups,and the incidence of injury was 100%.On the3 d,7d,the proportion of injury score of 2 in the middle and high concentration groups was significantly higher than that in the 5%GS control group and the low solubility amiodarone group,and there were significant differences among the three groups(P<0.05).After subcutaneous injection of solution for half an hour,3 d and 7 d,the injury occurred in all groups,and the smallest injury area were in the 5% GS control group,which was significantly different from that in the low,middle and high concentration amiodarone groups(P<0.05).After subcutaneous injection of the solution for half an hour,pathological changes occurred in the skin tissue structure.On day 7,the skin tissue structure was normal in the 5% GS control group,and there were still inflammatory cell infiltration in the dermis of the medium and high concentration amiodarone groups.2.After amiodarone extravasated,the levels of IL-6 and TNF-α in the tissue of each group increased rapidly,reached the peak on the 4th day and then decreased.The histological changes in the high chrysin dose intervention group were lower than those in the control group and10%DMSO solvent group.On day 3,7 and 10 of administration,the injury area and level of IL-6 and TNF-α in the chrysin groups were lower,when compared to the control group and 10% 10% DMSO solvent control group(P < 0.05),while the protein expression of b FGF on day 3,7 were higher in the chrysin middle-dose and high-dose groups,when compared to the control group and 10% DMSO solvent control group(P < 0.05).Conclusion:1.Amiodarone extravasation will cause histopathological changes and skin damage.The higher the concentration of amiodarone,the more serious the injury skin injury happened.2.Early intervention with chrysin can reduce the injury degree of amiodarone extravasation wound,reduce the infiltration of inflammatory cells in tissue,control the inflammatory response of wound by inhibiting the excessive secretion of TNF-α and IL-6 during cell proliferation,and stimulate tissue secretion of b FGF,to promote fibroblast proliferation and collagen synthesis,promote granulation tissue production,and then promote wound healing.
