The leaves of Crataegus pinnatifida Bge.are the dried leaves of Crataegus pinnatifida Bge.var.major N.E.Br.or Crataegus pinnatifida Bge.In this thesis,the chemical components contained in Crataegus pinnatifida Bge.leaves were summarized,and the active components,disease-related target proteins and signal pathways that could fight hyperlipidemia in Crataegus pinnatifida Bge.leaves were preliminarily predicted and explored by using network pharmacological research methods.Combined with the compounds isolated from Crataegus pinnatifida Bge.leaves in the experimental group,the therapeutic potential of chemical components in Crataegus pinnatifida Bge.leaves for hyperlipidemia was verified through the detection of ERK2 kinase activity and the establishment of high-fat Hep G2 cell model.1、Using the network pharmacological research method,this paper explored the active ingredients,key targets and related action pathways in Crataegus pinnatifida Bge.leaves with potential efficacy against hyperlipidemia.Firstly,the Crataegus pinnatifida Bge.leaves compound database and the target protein database related to hyperlipidemia were established,and the signal pathways related to the targets were collected.Secondly,molecular docking technology was used for preliminary screening,and compound-target protein network chart,PPI network chart,GO and KEGG enrichment analysis chart and compound-target-pathway network chart were constructed.Several key anti-hyperlipidemia compounds,targets,and signaling pathways were inferred from these analysis.2、In this paper,six compounds isolated from Crataegus pinnatifida Bge.leaves in the experimental group were used to test the activity of ERK2 kinase.The results showed that some of the compounds had certain inhibitory effects on ERK2 kinase.Among them,the highest inhibitory rate of vitexin on ERK2 kinase was the highest among all the compounds,with an inhibition rate of 84%and an IC50 value of 20.422μM.In this study,a high-fat Hep G2 cell model was successfully constructed,Oil Red O staining and quantitative experiments were carried out,and TG content was determined.It was found that with the intervention of vitexin,the number of lipid droplets in cells decreased gradually with the increase of vitexin concentration,and the triglyceride content decreased,indicating that vitexin can effectively reduce lipid accumulation and TG content in cells.Therefore,the results of network pharmacology in predicting the anti-hyperlipidemia effect of vitexin,a compound in Crataegus pinnatifida Bge.leaves,were verified.
