| Objective:To observe the effects of Yiyi Fuzi Baijiang San on uterine histomorphology and hemorheology in sequelae of pelvic inflammatory disease(SPID)model rats,and serum c-reactive protein(CRP),interleukin-2(IL-2),interleukin-10(IL-10),tumor necrosis factor alpha(TNF-α),malondialdehyde(MDA),superoxide dismutase(SOD)changes of the levels,and the expression of JNK and p38 signaling pathways that mediate the inflammatory response,to explore its possible mechanism of action in the treatment of SPID.Methods:60 SPF SD female rats were selected as experimental subjects,ten for the normal group,and the rest of the SPID rat models were prepared with the method of mixed bacterial suspension combined with instrument injury.After 30 days of modeling,the model rats were randomly divided into five groups: the model group,the high-dose group of Yiyi Fuzi Baijiang San,the medium-dose group of Yiyi Fuzi Baijiang San and the low-dose group of Yiyi Fuzi Baijiang San(hereinafter referred to as the high-dose group,the medium-dose group and the low-dose group),and the Fuke Qianjin Tablet group.After 21 days of intragastric administration,blood was collected from the abdominal aorta and bilateral uteri were taken.The uterine tissue morphology of rats was observed by HE staining.Meanwhile,the hemorheological indices of rats were detected by automatic hemorheological analyzer.The contents of CRP,IL-2,IL-10,TNF-α,SOD and MDA in the serum were determined by enzyme-linked immunosorbent assay(ELISA).Additionally,WB and PCR were used to detect the expression levels of JNK and p38 signaling pathways in rat uterine tissues.Result:1.Morphological changes of rat uterine tissue: The physiological structure of the uterus in the normal group was complete,without inflammation,edema or other pathological changes.In the model group,there were obvious inflammatory cell infiltration and vascular congestion in the uterine mucous layer,lamina propria and the interstitial cells.Different degrees of inflammatory cell infiltration and vascular congestion were observed in the medium and low dose groups.Moreover,Uterine physiological structure of rats in the high-dose group and the Fuke Qianjin Tablet group was basically normal,and no inflammatory cell infiltration was observed in the mucous layer,lamina propria and the interstitial cells.2.Changes of hemorheological indexes: Compared with the normal group,the hemorheological indices of rats in the model group were significantly increased(P<0.05).Compared with model group,the hemorheological indices of rats in the high-dose and medium-dose groups and the Fuke Qianjin Tablet groups significantly decreased(P<0.05).3.Changes in levels of inflammatory cytokines: Compared with normal group,serum levels of CRP,IL-2 and TNF-α in the model group significantly increased,and the IL-10 levelsignificantly decreased(P<0.05).Compared with model group,the serum levels of CRP,IL-2 and TNF-α in the high-dose,medium-dose and low-dose groups and the Fuke Qianjin Tablet groups significantly decreased,while the level of IL-10 significantly increased(P <0.05).4.Changes of oxidative stress indices: Compared with the normal group,the MDA level in the serum of the model group significantly increased,and the SOD level significantly decreased(P<0.05).Compared with model group,the MDA level in the serum of rats in the high-dose,medium-dose and low-dose groups and the Fuke Qianjin Tablet groups were significantly decreased,while the SOD level significantly increased(P<0.05).5.WB and PCR methods were applied to detect the expressions of JNK and p38 signaling pathways: Compared with the normal group,the expression levels of JNK and p38 signaling pathways in the uterine tissues of rats in the model groupsignificantly increased(P<0.05).Compared with model group,the expression levels of JNK and P38 signaling pathways in the uterine tissues of rats in the high-dose and medium-dose groups and the Fuke Qianjin Tablets groups significantly decreased(P<0.05).Conclusion:The results of this study demonstrated that Yiyi Fuzi Baijiang San could significantly improved the pathological morphology of uterus tissue in SPID model rats.It also reduced various hemorheological indices,effectively regulated the levels of CRP,IL-2,IL-10,TNF-α,SOD and MDA,and inhibited the expressions of JNK and P38 signaling pathways.Consequently,it can be inferred that Yiyi Fuzi Baijiang San may achieve the therapeutic effect of SPID by improving hemorheological indices and reducing the level of pro-inflammatory factors,increasing the level of anti-inflammatory factors,regulating the balance of the body’s oxidation-antioxidation systemand,inhibiting the expression of JNK and P38 signaling pathways that mediate the inflammatory response. |
