Previous literatures have showed that polyunsaturated fatty acids have antineoplastic activities, but the mechanisms are still unknown exactly. In the present study, related indicators have been estimated including level of reactive oxygen species, activity of antioxidant enzyme, accumulation of Iipid peroxides and content of lipoxin, along with changes of fatty acid composition in the process of intracellular fatty acid de novo synthesis and unsaturated fatty acid metabolism in both gastric normal and cancer cells treated by PUFAs. Results of this investigation are as follows:(1) DHA inhibited proliferation and induced apoptosis of gastric cancer cells significantly but protected stomach normal cell from injury, while to a’certain extent LA, ALA, AA and EPA all inhibited proliferation and induced apoptosis of gastric cancer cells meanwhile exerted great effect on stomach normal cell;(2) Further researches showed that there existed close internal relationships among lipid peroxidation, de novo synthesis of fatty acids, PUFAs metabolism and the contribution of PUFAs on cell-growth. In conclusion, DHA protected GES1cell from injury through promoting the absorption amount of the cell on itself and the promotion synthesis of oxygenated derivative LXA4. Differently, other PUFAs impaired GES1cell due to the large amount of PUFAs uptake into the cell and their serious lipid peroxidation and also might result from their competitive inhibition of fatty acids de novo synthesis. Otherwise, that PUFAs influence gastric cancer cells growth could attribute to a high degree of their peroxidation and a great decrease in their active metabolites.
