ObjectiveThe aim of this study was to observe the hypoglycemic effect of Sanye Table on healthy subjects after taking starch,and investigate the alleviating of Sanye Table on the whole process of starch decomposition in vitro conditions,and then analyze the effects of sanye table on variousα-glycosidase enzymes involved in starch hydrolysis.To find the way to interrupter starch hydrolysis by Sanye Tablet and provide support for the mechanism of Sanye Tablet.Methods1.In this study,a two-cycle self-cross-control design was adopted,and 12 healthy subjects were selected to receive two intervention methods:starch aqueous solution in the first cycle and starch aqueous solution in the second cycle plus Sanye Tablet,to observe the effects of Sanye Tablet on postprandial blood glucose and blood glucose fluctuations of healthy people after taking starch.2.Using starch,dextrin,maltose and isomaltose as substrates,the inhibitory effects of Sanye Tablet onα-glycosidase(amylase,dextrinase,maltase and isomaltase)in starch hydrolysis was investigated.Results:1.Clinical research:(1)The peak of postprandial blood glucose in starch and starch+Sanye Tablet occurred at 45min postprandial.(2)Sanye Tablet can reduce the blood glucose level of healthy people at each time point after taking starch,and there was statistical significance in the decrease of blood glucose at 5time points after taking starch at 15min,30min,45min,60min and 90min(P<0.05).There was no significant difference in the blood glucose level at 120min after starch administration(P>0.05).(3)Sanye Tablet can slow down the fluctuation of blood glucose in healthy people after taking starch.The fluctuation value of blood glucose in the starch+Sanye Tablet group is0.82 mmol/L,and the fluctuation value of blood glucose in the starch group is 1.33 mmol/L.2.In vitro research:(1)In the concentration range of 6.88×10-5mg/ml to 5.50×10-4mg/m;,Sanye Tablet had no obvious inhibitory effect on amylase activity.(2)In the concentration range of 1.10×10-4mg/ml to 6.60×10-3mg/ml,Sanye Tablet had no significant inhibitory effect on dextrinase activity.(3)In the concentration range of 5.50×10-4mg/ml to 1.10×10-2mg/ml,Sanye Tablet had a significant inhibitory effect on maltase activity,and the IC50value was(3.041±0.038)×10-3mg/ml.(4)In the concentration range of 5.50×10-4mg/ml to 1.10×10-2mg/ml,Sanye Tablet had a significant inhibitory effect on isomaltase activity,and the IC50value was(1.721±0.013)×10-3mg/ml.Conclusion:1.The results of clinical studies have concluded that the Sanye Tablet can reduce the postprandial blood glucose level and slow down the fluctuation of blood glucose in healthy people after taking starch.2.The results of in vitro inhibition study showed that Sanye Tablet have no obvious inhibitory effect on amylase and dextrinase activities,but have a good inhibitory effect on maltase and isomaltase activities.3.Hypoglycemic effect of Sanye Tablet was achieved by inhibiting the activity of diglyase downstream of starch hydrolysis,but not directly inhibiting the activity of amylase.4.The traditional Chinese medicine compound Sanye Tablet is a potentialα-glycosidase inhibitor.
