Effect Of Heat-sensitive Moxibustion Intervention On Neuroinflammation In Alzheimer’s Disease Rat

Objective:To observe the effect of heat-sensitive moxibustion on the levels of IL‐1β,IL‐6,TNF‐αand NF‐кB in the serum of Aizheimer’s disease(AD)rat models.In order to explore the curative effect and mechanism of heat-sensitive moxibustion in the treatment of AD.Method:86 SD rats meeting the inclusion criteria will be divided into operation group(70),sham operation group(8)and control group(8)randomly.Among them,the successfully prepared rats were evaluated by the water maze experiment,and then randomly divided into model group(9),moxibustion treatment group(52),and positive control group(9).The positive control group received the drug donepezil intervention,0.5mg/kg/d;the moxibustion treatment group received moxibustion treatment at the "Dazhui point" on the neck of the rat,40min/time,once a day.According to the change of tail temperature during moxibustion,it was divided into three subgroups: heat-sensitive moxibustion(HSM)group,delayed heat-sensitive moxibustion(DHSM)group,and non-heat-sensitive moxibustion(NHSM)group.The other groups received no other treatment.After 28 days of treatment,the behavioral changes,the brain histomorphology changes,and the content of IL-1β、IL-6、TNF-α、NF-κB in serum of rat in each group will be used as our evaluation indicators.Result: Results of the water maze :After successful modeling,the escape latency of AD rats on day 1-5 was significantly increased(P<0.01),and the times of crossing the platform and ratio of residence time was significantly reduced(P<0.01).After the intervention of heat-sensitive moxibustion and donepezil,it can significantly shorten the escape latency of AD rats on day 1-5(P<0.01),and increase the times of crossing the platform and ratio of residence time(P <0.01).Among the three subgroups,the effect of HSM group was better.The escape latency on day 1-5 of rats was significantly decreased(P<0.05),the times of crossing platform and ratio of residence time were significantly increased(P<0.05).Results of the HE staining: The hippocampal neurons and glial cells of the control group and the sham-operated group were arranged orderly with normal structure,the cytoplasm was abundant,and no necrosis.In the model group,the brain tissue was seriously damaged,a large number of neurons were lost and apoptotic,the arrangement of nerve cells was disordered,the cell morphology was abnormal and swollen,and some nuclei were dissolved and pyknosis.Compared with the model group,the brain tissue injury was reduced of all treatment groups.The HSM group had the lowest degree of damage,the brain tissue morphology was relatively normal,the cells were arranged in order,and there was no aggregation of inflammatory cell.HE semi-quantitative results:Compared with the model group,the HE semi-quantitative scores of rats in the other treatment groups decreased significantly(P<0.05 or P<0.01),except for the NHSM group.Among the three subgroups,the intervention effects of HSM group and DHSM group were better(P<0.05).Results of the ELISA :The contents of IL-1β,IL-6,TNF-αand NF-κB in serum of rats after successful modeling were significantly increased(P<0.01).After heat-sensitive moxibustion and donepezil intervention,the contents of IL-1β,IL-6,TNF-αand NF-κB in serum were all decreased in varying degrees(P<0.05 or P<0.01).Among the three subgroups,the HSM group had better effect,and the contents of IL-1β,IL-6,TNF-αand NF-κB were significantly decreased(P<0.05 or P<0.01).Conclusion:1 Heat-sensitive moxibustion can effectively improve the spatial learning and memory ability of AD rats,and the effect of rapid heatsensitization of acupoints is better;2 Heat-sensitive moxibustion can effectively reduce the brain tissue necrosis of AD rats;3 Heat-sensitive moxibustion can significantly reduce the levels of IL-1β,IL-6,TNF-α,NF-κB in brain tissue of AD rats,and the intervention effect of rapid heat-sensitization of acupoints is more effective than that of delayed type,which may be one of the mechanisms of heat-sensitive moxibustion therapy plays a role in protecting neurons.
