Background Cervical cancer is one of the most common malignant tumors of female reproductive system.Persistent infection of HPV is closely related to the occurrence of cervical cancer.With the development of molecular biology,the occurrence of cervical cancer may involve the role of other oncogenes.Objective To study the expression and clinical significance of FoxM1 and SIRT1 in cervical benign lesions,low-grade squamous intraepithelial lesions(LSIL),high-grade squamous intraepithelial lesions(HSIL)and cervical squamous cell carcinoma(CSCC),and to explore the relationship between the two genes in LSIL,HSIL and CSCC tissues,so as to provide some basis for the study of pathogenesis and molecular targeted therapy of cervical cancer.Methods The cervical tissue samples of 140 patients in the department of gynecology and oncology in our hospital from January 2015 to January 2019 were collected,and the clinicopathological parameters were recorded.All the specimens were confirmed by pathology.There were 30 cases of cervical benign lesions,30 cases of LSIL,30 cases of HSIL and 50 cases of CSCC.Immunohistochemical method was used to detect the expression of FoxM1 and SIRT1 in cervical benign lesions,LSIL,HSIL and CSCC tissues.Combined with clinicopathological parameters,statistical software was used to analyze the significance of the expression of the two genes in cervical squamous cell carcinoma and their correlation in LSIL,HSIL and CSCC tissues.Results(1)The positive rate of FoxM1 expression in cervical benign lesions,LSIL,HSIL and CSCC tissues gradually increased,which was 13.33%,53.33%,76.67% and 90.00%,respectively.There was significant difference among cervical benign lesions,SIL and CSCC tissues(P < 0.01).(2)The positive rate of SIRT1 expression in cervical benign lesions,LSIL,HSIL and CSCC tissues showed a gradually increasing trend,which was 0.00%,56.67%,80.00% and 92.00%,respectively.There was significant difference among cervical benign lesions,SIL and CSCC tissues(P < 0.01).(3)There was no clear difference in the positive expression of FoxM1 and SIRT1 in CSCC tissues in terms of clinical stage,degree of differentiation,lymph node metastasis,vascular invasion and myometrial invasion(P > 0.05).(4)There was a definite positive correlation between the expression of FoxM1 protein and SIRT1 protein in LSIL,HSIL and CSCC tissues(P < 0.01).Conclusion(1)FoxM1 and SIRT1 may be involved in the development of cervical benign lesions to precancerous lesions and cervical cancer.Detection of their expression in patients with cervical lesions may be helpful to detect the early lesions of cervical cancer.(2)The expression of FoxM1 and SIRT1 protein in CSCC was not related to clinical stage,degree of differentiation,lymph node metastasis,vascular invasion and myometrial invasion.(3)There may be some correlation between FoxM1 and SIRT1 proteins in LSIL,HSIL and CSCC,but the specific relationship and mechanism need to be clarified by further experimental study.
