| Immunoassay plays an important role in clinical detection.The immunoassay method based on inductively coupled plasma mass spectrometry(ICP-MS)has the advantages of low matrix effect,high sensitivity and multi-component simultaneous detection.At present,in the field of ICP-MS immunoassay,nanoparticles are the best probe for signal amplification,and the types of nanoparticles and the detection conditions of instruments need to be further explored.In this paper,we use ferritin as the model,synthesized platinum nanoparticles as the probe,selected immunomagnetic spheres as the reaction and separation carrier,and used two different detection modes(signal intensity integration mode and single particle mode)of ICP-MS to establish two high-sensitivity immunoassay methods,which provides the theory and method for the establishment of immunoassay based on ICP-MS in the future.This method has potential application in the early diagnosis of clinical tumors and proteomics.The main research results are as follows:(1)Based on the signal intensity integration model,a method of nanoparticle labeled magnetosphere immunoassay was established for ICP-MS.A kind of porous platinum nanoparticle probe was synthesized.Through the optimization of immune reaction conditions and the adjustment of instrument parameters,the high sensitivity detection of ferritin with low concentration,which is of great clinical significance,was realized.The detection limit is 88 pg/m L,and the detection range is 1.25~250 ng/m L.the method has been proved to be selective,repeatable and consistency with clinical methods.Compared with other magnetic immunoassay,this method does not need dissociation step,is more convenient and efficient.(2)Based on the single particle model ICP-MS(sp-icp-ms),a nano platinum labeled magnetic immunoassay was established.The sp-icp-ms detection method of platinum nanoparticles was explored,the sample transfer efficiency and injection method were optimized based on the principle of the instrument,and the dissociation method of immune complex was optimized.The most sensitive detection of ferritin in the reported literature was realized,the detection limit was 48 pg / m L,and the detection range was 1.25~250 ng/m L,which also passed the performance test of the actual samples.This method has potential for clinical application. |
