The dried rhizome from Acori Tatarinowii Schott is utilized as a traditional Chinese medicine,which is belongs to the Araceae family and is recorded in the Chinese Pharmacopoeia,processing memory enhancing property.It is containing volatile oil which including α/β/γ-asarone,calamusenone,elemene and so on.But,the non-volatile content is also having the pharmacological effects and like the part of water-soluble fraction was also eat for diseases treatment.Besides the value of pharmacological,ATR is used as the original material of food,like ChangPu Jiu,a Chinese spirit which is promoting blood circulation,enhancing memory and improving eyesight according to the book recorded from Han dynasty.However,there are two genu herbs(Acori Calami Rhizoma(ACR)and Anemones Altaicae Rhizoma(AAR))authented with ATR,sharing several morphological similarities,as well as have similar bioactivities for memory enhancing.Therefore,two analysis platforms,Ultra High Performance Liquid Chromatography-High Resolution Mass Spectrometry(UPLC-MS/MS)and Inductively Coupled Plasma Mass Spectrometry(ICP-MS)were applied to analyze and explore the ATR and its adulterants,as well as to study the changes of before and after fermentation,in this study.The main research contents can be divided to such aspects as follows:1.Analysis of organic chemical components in ATR,ACR and AAR.27 batches of ATR,13 batches of ACR and 14 batches of AAR were extracted with 70% methanol water and analyzed by UPLC-Orbitrap-MS/MS.55 organic components were identified from ATR and ACR;36 organic components were from AAR based on the retention time and information of calculated molecular ion and major fragments,as well as the fragmentation pathways.Magnolol was used as the internal standard for semi-quantitating.The results of Duncan’ test show that 32 of the 36 common compounds were found to be different among ATR,ACR and AAR.Studies have shown that there were significant differences in organic compositions between AAR and ACR,ATR.2.Analysis of Chemical Elements in ATR,ACR and AAR.27 batches of ATR,13 batches of ACR and 14 batches of AAR were digested with nitric acid and perchloric acid in an automated digestion system DEENAⅡ(Thomas Cain,USA)and analyzed by ICP-MS.Standard curve,precision and recovery rate were analyzed in order to ensure the quality of the experimental data.40 elements were detected and quantitative.The results showed that almost all 40 elements have exhibited significant differences between AAR and ATR.And the difference between AAR and ATR is greater than that between ACR and ATR in elements.3.Differential analysis on the chemical components of ATR,ACR and AAR.Discriminant analysis was applied based on the analyzed combined with UPLC-OrbitrapMS/MS and ICP-MS.β-asarone,can distinguish AAR from ATR and ACR according to the scores’ plot and loading’s plot obtained by principal component analysis(PCA).Partial least squares-discriminant analysis(PLS-DA)coupled with two variable selection methods(VIP,coefficient β)was used to further discriminate analysis of ATR and ACR.The model,VIP-PLS-DA,have a perfect result of discriminant.After variable selection by VIP,twenty-one chemical markers were identified for the discrimination between ACR and ATR and used to establish their characteristic quantitative fingerprints.4.Study on the Changes of Organic Chemical Components in ATR during fermentation.The fermentation liquid of ATR in 72,96,120 and 144 hours was detected by UPLC-MS / MS.Seven compounds,salicylic acid,6,7-dihydroxycoumarin,caffeic acid,1-cis-propenyl-1S,6R-epoxy-4-methoxy-2,5-quinone,chlorogenic acid,tatarine C,significantly increased,while phenylalanine and tatarinoids B was significantly reduced in the process of fermentation.
