| Objective:1.To detect the expression of never in mitosis gene A related kinase-2(NEK2)and P53 in normal cervical tissues and cervical squamous cell carcinoma(CSCC)tissues.2.To analyze the relationship between the expression of NEK2 and the age,tumor size,histological differentiation,and lymph node metastasis of cervical cancer patients.3.To analyze the correlations between NEK2 and P53 expressions in cervical cancer tissues.4.To analyze the expression levels of NEK2 and multidrug resistance-associated protein(MRP)in Si Ha and Si Ha/VP16 cells.Methods:1.Selected 40 cases paraffin specimens of cervical cancer that were surgically removed from the gynecological ward of the First Hospital of Shanxi Medical University from August 2018 to December 2020 as the experimental group,and 40 cases paraffin specimens of normal cervical tissues that were derived from cervix undergoing total hysterectomy due to uterine fibroids or pelvic organ dysfunction diseases as the control group.All specimens were clearly diagnosed by pathological examination.2.Immunohistochemistry(IHC)was used to detect the expressions of NEK2 and P53 in the two groups of specimens.3.Analyzed the relationship between NEK2 expression and clinical pathological factors such as the age,tumor size,histological differentiation,and lymph node metastasis of cervical cancer patients.4.Analyzed the correlations between NEK2 and P53 expressions in cervical cancer tissues.5.Cervical cancer Si Ha cells and multidrug resistant Si Ha/VP16 cells were cultured in vitro,and the expression levels of NEK2 and MRP protein in Si Ha and Si Ha/VP16 cells were detected by westrn blotting.Results:1.The results of immunohistochemisty showed that the positive expression rates of NEK2 in normal cervical tissues and CSCC tissues were 22.50%(9/40)and 67.50%(27/40)respectively.The expression of NEK2 in CSCC tissues was significantly higher than that in normal cervical tissues.The difference was statistically significant(χ~2=16.364,P=0.000).2.The results of immunohistochemisty showed that the positive expression rates of P53 in normal cervical tissues and CSCC tissues were 10.00%(4/40)and 47.50%(19/40)respectively.The expression of P53 in CSCC tissues was significantly higher than that in normal cervical tissues.The difference was statistically significant(χ~2=13.730,P=0.000).3.The positive expression rates of NEK2 in high,medium,and poorly differentiated CSCC tissues were 20.00%(1/5),70.83%(17/24),81.82%(9/11),respectively.The expression of NEK2 in poorly differentiated CSCC tissues was significantly increased,and the difference was statistically significant(χ~2=6.292,P=0.043).The positive expression rate of NEK2 in CSCC tissues without lymph node metastasis was 60.61%(20/33)and in CSCC tissues with lymph node metastasis was100.00%(7/7).The expression of NEK2 in CSCC tissues with lymph node metastasis was significantly increased,and the difference was statistically significant(χ~2=4.058,P=0.043).There was no statistically significance between the expression of NEK2 and the age of patients,and tumor size(P>0.05).4.Pearson correlation analysis showed that the expressions of NEK2 and P53 were positively correlated in CSCC tissues.The correlation coefficient was 0.553,and P=0.000.5.The results of western blotting showed that the relative expressions of NEK2 in Si Ha and Si Ha/VP16 cells were 0.5397±0.1314 and 1.0013±0.2146,respectively.Compared with Si Ha,the expressions of NEK2 in Si Ha/VP16 cells were significantly higher,and the difference was statistically significant(t=-3.178,P=0.034).The relative expressions of MRP in Si Ha and Si Ha/VP16 cells were 0.3160±0.1543 and0.4803±0.1831,respectively.Compared with Si Ha,the expressions of MRP in Si Ha/VP16 cells were significantly higher,and the difference was statistically significant(t=-1.189,P=0.030).Conclusion:1.NEK2 is highly expressed in CSCC and is related to the degree of tumor differentiation and lymph node metastasis.It may participate in the pathological process of the occurrence and development of CSCC,and is expected to become an effective gene target for CSCC research.2.The expressions of NEK2 and P53 in CSCC are positively correlated.NEK2may play a role in the progression of CSCC through the molecular pathway of P53 gene.3.The expression of NEK2 in cervical cancer multidrug resistance cell lines is significantly increased,which may be related to the chemotherapy resistance of cervical cancer. |
