Sodium Aescinate Inhibits The Proliferation Of Small Bile Ducts In Rats With Obstructive Jaundice By Reducing The Phosphorylation Level Of 4EBP1

Abstract/Summary:
Objective In order to investigate the expression and biological relationship of 4EBP1 in the liver tissue of rats with obstructive jaundice and to study the inhibitory effect of sodium aescinate on the proliferation of bile duct and the improvement of cholestatic liver fibrosis in rats with obstructive jaundice.Methods 81 SD rats were divided into bile duct ligation group(OJ),bile duct ligation +Sodium Aescinate treatment group(SA)and sham operation group(Sham).The rats in SA group were given sodium aescinate on the first day after operation,and the rats in OJ group and Sham group were given corresponding amount of normal saline to ensure the same feeding environment of the three groups.After 7 days of continuous administration,the rats were killed,blood samples were taken from inferior vena cava and liver samples were obtained.HE staining,Masson staining,immunofluorescence staining and immunohistochemical staining were used to observe the liver tissue.The staining of liver tissue of rats in each group was taken by microscope,and then the images were analyzed by Image Pro Plus 6.0 software to obtain the experimental data.SPSS 22.0 software was used to analyze and process the experimental data.Results(1)48 hours after bile duct ligation,the urine color of rats in OJ group and SA group was yellow,the ears,eyes,hair and skin were yellow,and the faeces were light gray;In Sham group,the urine was light yellow,but the ears,eyes,hair and skin were not yellow.(2)The results of HE staining showed that the radiated hepatic cords were visible in Sham group,no obvious connective tissue hyperplasia was found,and the morphology and structure of liver were normal.No hyperplastic bile duct was found in the portal area of liver tissue,and no cholestatic fibrosis was found in the liver;In OJ group,the structure of hepatic cord was disordered,the space expanded and the vacuoles increased.A large area of necrosis of liver tissue could be seen.The whole microscopic field was full of inflammatory cells,and a large number of small bile ducts could be seen around the portal area;In SA group,the hepatic cords were regular,and a few vacuoles could be seen under the microscope.Compared with OJ group,the number of bile ducts around the portal area of liver tissue was significantly reduced,and the structural changes of hepatic lobules were also significantly improved.(3)Masson staining showed that compared with sham group,the area and average collagen density of liver collagen fibers in OJ group were significantly increased(P<0.05);Compared with OJ group,the area and density of hepatic collagen fibers in SA group were significantly changed(P<0.05).(4)The results of immunofluorescence showed that the contents of COL-Ⅰ and COL-Ⅲ in liver of Sham group were the least,and the contents of COL-Ⅰ and COL-Ⅲ in liver of OJ group were significantly higher than those of Sham group(P<0.05);Compared with OJ group,the protein contents of COL-Ⅰ and COL-Ⅲin liver of SA group were significantly decreased(P<0.05).(5)The results of immunohistochemical staining showed that there was no significant difference between sham group and Sham group the expression of α-SMA was the lowest and the positive staining area was the least;In OJ group,there was no significant difference between the two groups the expression of α-SMA was the highest,and the positive staining area was the largest;In SA group,there was no significant difference compared with the OJ group,the expression level of α-SMA decreased significantly,and the positive staining area decreased significantly.In Sham operation group,there was no positive expression of 4EBP1 and p-4EBP1 around the portal area;Compared with sham group,a large number of bile ducts were found in the liver of OJ group,and 4EBP1 and p-4EBP1 were significantly expressed in the columnar epithelial cells of bile duct.In SA group,there were only a few bile ducts around the hepatic portal area,and 4EBP1 was positive in bile duct cells,but p-4EBP1 was not positive in SA group.Conclusions Obstructive jaundice model rats caused cholestatic hepatic fibrosis,and a large number of bile ducts around the portal area compensated.Sodium aescinate can inhibit the proliferation of small bile duct in rats with obstructive jaundice by reducing the expression of p-4EBP1 in bile duct columnar epithelial cells.
