| Objective:This experimental study aims to observe the effects of Compound Qinglongyi Capsule on 24h-UPr,renal function and some biochemical indexes of liver function,IL-6 and TGF-β1 in rats with adriamycin nephropathy from the perspective of inflammatory response.The feasibility of the capsule in the treatment of proteinuria in rats with adriamycin-induced nephropathy provides a theoretical basis for exploring the new efficacy of the drug.Methods:In the experiment,72 6-week-old clean-grade male SD rats were selected.After one week of adaptive feeding,they were first randomly divided into two groups according to the weight of the rats,namely the blank group(n=12)and the model group(n=60).According to the Bertani method,the model rats were established by a one-time tail vein injection of adriamycin solution at a dose of 6.5 mg/kg body weight.The modeled mice24h-UPr≥10mg and statistically significant were used as the criterion for successful model building.The successfully modeled and surviving rats were randomly divided into four groups,namely the compound Qinglongyi group(Qinglongyi group),Tripterygium wilfordii polyglycosides group(tripterygium wilfordii),and Benazepril hydrochloride tablets group(Luotensin group)),model control group(model group).Observe and record the general state and related data of rats in each group on time every day.After successful modeling,the 24h-UPr of rats was measured on the weekends 1,2,and 4.At the end of the 4th week,the rats were intraperitoneally injected with 10% chloral hydrate solution,and blood was taken from the heart after anesthesia to detect Scr,BUN,TC,ALB,ALT and AST.Cut the kidney tissue in a coronal plane,and prepare a 10%homogenate with a homogenizer under ice-water bath conditions;the other part is fixed in 2.5% glutaraldehyde to prepare for electron microscopy to observe the changes in renal tissue structure;Immunohistochemical staining method was used to analyze the localization and expression of IL-6 and TGF-β1 in kidney tissue;PCR detection method was used to determine the content of IL-6 and TGF-β1 in kidney tissue.Results: 1.The new dragon clothing,the new group of Lotin and the24h-UPr of the Rayhagi rats were reduced(P<0.05);the cyano group rat,compared with the new group of 24h-UPr significantly reduced(P<0.05),compared with the Rayhavi group,there is no significant difference(P> 0.05;2.New dragon clothing,the new group of Lotin and the rat value of the Rayhais rats,the model group,Qinglong clothes The group was significantly reduced(P<0.05);the rats in the cyano group and the Lotin group were compared,the Tc value was significantly reduced(P<0.05).Qinglong Clothing,the new group of Lotin and the rats of the Rayhais rats increased by the model group,the rise in the Qinglong Clothing Group is the most obvious(P<0.05);Youth Legal Group Rats and Rayliha Group,Lotin The new group compared,the ALB value was significantly increased(P<0.05);3.The new dragon clothing,the new group of losin and the rats in rats in rats in rats,and the expression of TGF-β1 in the model group The decrease is reduced,and the difference has statistically significant(P<0.05);4.Transmission electron microscopic results showed that the ghostadecoelectric foot cycle integration even disappeared in the Qinglong coat,Radiate vine group rats.Conclusion:1.Compound Qinglong Capsules can effectively reduce the quantification and serum total cholesterol content of doxorubic nephropathy rats,raising serum albumin content,slowing further renal damage,thereby delays the development of kidney disease,and play a certain kidney protection Role;2.Compound Qinglong Capsule may reduce the damage of kidney inflammatory response by reducing the expression and quantification of IL-6,TGF-β 1,reducing the damage of kidney inflammatory response,thereby inhibiting proteinuria. |
