Objective: To explore the role of pyroptosis in the pathophysiological changes of bladder caused by bladder outlet obstruction(BOO)in mice.Preliminarily explore the effect of BOO on cell surface integrins and its relationship with pyroptosis.To further understand and understand the characteristics of changes in the function of bladder smooth muscle during BOO.To find new targets for the prevention and treatment of bladder dysfunction after BOO.Methods: The mouse model of bladder outlet obstruction was established artificially,and the expression of protein and gene in bladder was detected by Western blotting and q RTPCR.Histomorphological changes of bladder were observed by H&E and Masson’s trichrome staining.We reversely verified the effects of pyroptosis,Integrin α5 and CD47 on the BOO bladder by giving mice NLRP3 inhibitor Glyburide,Integrin α5 inhibitor ATN-161,and CD47 inhibitor RRX-001.Results: Compared with the control group,the bladder of the mice in the BOO group showed stronger pyroptosis and inflammation,which gradually aggravated with the prolongation of the obstruction time.The expression of integrin α5 and CD47 increased significantly after BOO.Glyburide and ATN-161 can significantly inhibit the progression of pyroptosis and the thickening of the bladder wall.The CD47 inhibitor RRX-001 only showed an effect on bladder remodeling and had no effect on pyroptosis.Conclusion: Pyroptosis plays an important role in the damage of bladder function caused by BOO.Integrin α5 and CD47 are also involved in signal transmission in BOO.Inhibitors for integrin α5,CD47,and NLRP3 inflammasome have shown to significantly delay BOO bladder remodeling,which may provide a new treatment strategy for BOO drug treatment.
