| Objective:To observe the effects of glutathione(GSH)on the contents of malondialdehyde(MDA),adenosine triphosphate(ATP),and the activity of glutathione peroxidase(GSH-PX)and superoxide dismutase(SOD)in stored red blood cells(sRBC)at different times after storage.Methods:The prepared sRBC 12 ml were divided into 4 groups:group ACD,group GSH1,group GSH2 and group GSH3.GSH 50.0μl was added into the sRBC in group GSH1,group GSH2 and group GSH3,and the final concentration of GSH was controied in12.5,62.5 and 125μmol/L respectively,But it was only 50.0μl normal saline added in the group ACD.All of the sRBC of the 4 groups were stored in the refrigerator at(4±2)℃.On the 1st day,7th day,14th day and 21st day of preservation,the contents of ATP,MDA,and the activity of SOD and GSH-PX in the four groups of sRBC were determined.and the osmotic pressure,pH value,concentration of K~+、Na~+,and glucose and lactic acid of the preservation solution were measured at the same times.Results:1.The osmotic pressure,pH value,concentration of K~+、Na~+,glucose and lactic acid in the4 groups of sRBC preservation solution.Comparison between groups:there was no significant difference in osmotic pressure,pH value,concentration of K~+、Na~+,glucose and lactic acid in the 4 groups of sRBC preservation solution on the 1st,7th,14th and 21st day of preservation(P>0.05).Comparison within the group:there was no significant difference in the osmotic pressure of sRBC preservation solution among the 4 groups at each time point(P>0.05).The pH value of the preservation solution decreased gradually with the extension of preservation time in the 4 groups(after the 7th day,there was significant difference between each time point,P<0.05);The concentration of Na~+and glucose in sRBC preservation solution of the 4 groups decreased gradually with the extension of preservation time(after the 7th day,there was significant difference between each time point,P<0.05),while the concentration of K~+and lactic acid in sRBC preservation solution of the 4 groups increased gradually(after the 7th day,there was significant difference between each time point,P<0.05).2.The changes of ATP levels in 4 groups of sRBC.Comparison between groups:on the 1st,7th,14th and 21st day,the content of ATP in sRBC of group GSH2 and group GSH3 was higher than that of group ACD(P<0.05),and the content of ATP in sRBC of group GSH2and group GSH3 was higher than that of group GSH1(P<0.05).Comparison within the group:the content of ATP in sRBC of group ACD on the 21st and 14th day of storage was significantly lower than that on the 7th and 1st day(P<0.05),and the content of ATP in sRBC of group GSH1 and group GSH2 on the 21st day of storage was significantly lower than that on the 14th,7th and 1st day(P<0.05),and that on the 14th day was significantly lower than that on the 7th and 1st day(P<0.05).In group GSH3,the content of ATP in sRBC on the 21st day after storage was significantly lower than that on the 14th,7th and1st day(P<0.05),and that on the 14th day was significantly lower than that on the 7th and1st day(P<0.05),and that on the 7th day was significantly lower than that on the 1st day(P<0.05).3.The changes of MDA levels in 4 groups of sRBC.Comparison between groups:on the1st,7th,14th and 21st day of storage,the content of MDA in sRBC of group GSH2 and group GSH3 was lower than that of group ACD(P<0.05),and the content of MDA in sRBC of group GSH2 and group GSH3 was lower than that of group GSH1 on 1st,7th and21st day of storage(P<0.05).Comparison within the group:the content of MDA in sRBC of group ACD,group GSH1,group GSH2 and group GSH3 on the 21st day after storage was significantly higher than that on the 14th,7th and 1st day(P<0.05),and that on the14th day was significantly higher than that on the 7th and 1st day(P<0.05),and that on the 7th day was significantly higher than that on the 1st day(P<0.05).4.The changes of SOD enzyme activity in 4 groups of sRBC.Comparison between groups:on the 1st,7th,14th and 21st day,the activity of SOD in the sRBC of group GSH2 was higher than that of group ACD(P<0.05),and on the 1st,7th and 21th day,the activity of SOD in the sRBC of group GSH3 was higher than that of group ACD(P<0.05),and the activity of SOD in the sRBC of group GSH2 and group GSH3 was higher than that of group GSH1 on the 21st day(P<0.05).Comparison within the group:the activity of SOD in sRBC of group ACD and group GSH3 on the 21st day after storage was significantly lower than that on the 14th,7th and 1st day(P<0.05),and the activity of SOD in sRBC of group GSH2 and group GSH1 on the 21st day after storage was significantly lower than that on the 14th,7th and 1st day(P<0.05),and that on the 14th day was significantly lower than that on the 7th and 1st day(P<0.05),and that on the 7th day was significantly lower than that on the 1st day(P<0.05).5.The changes of GSH-PX enzyme activity in 4 groups of sRBC.Comparison between groups:on the 1st,7th,14th and 21st day,the activity of GSH-PX in sRBC of group GSH2and group GSH3 was higher than that of group ACD(P<0.05),and the activity of GSH-PX in sRBC of group GSH2 and group GSH3 was higher than that of group GSH1(P<0.05).Comparison within the group:the activity of GSH-PX in sRBC of group ACD,group GSH1,group GSH2 and group GSH3 on the 21st day after storage was significantly lower than that on the 14th,7th and 1st day(P<0.05),and that on the 14th day was significantly lower than that on the 7th and 1st day(P<0.05),and that on the 7th day was significantly lower than that on the 1st day(P<0.05).Conclusion:1.With the extension of storage time,the content of ATP,the activity of SOD and GSH-PX of the sRBC gradually decrease,and the content of MDA gradually increases.The change of these indexes could be alleviated by adding a certain concentration of GSH;2.With the prolongation of storage time,the pH value,the concentration of Na~+and glucose of the sRBC preservation solution gradually decrease,and the concentration of K~+,lactic acid gradually increase.Adding a certain concentration of GSH cannot change the trend of these indicators. |
