Objective:To investigate the safety and clinical efficacy of vitamin C(Vit C)combined with azithromycin in the adjuvant treatment of Mycoplasma pneumoniae pneumonia(MPP)in children,and to detect relevant experimental indicators to preliminaries explore the mechanism of action,in order to provide reference for clinical medication.Methods:100 cases of MPP children admitted to Zunyi maternal and child health hospital from July 2019 to September 2020 were selected and randomly divided into two groups(observation group and control group),50 cases in each group,At the same time,50healthy children were randomly selected as the healthy group.Azithromycin+vitamin C+routine treatment was the treatment plan of the observation group,and azithromycin+routine therapy was the control group’s treatment plan.The safety of Vit C was evaluated by monitoring the liver function,renal function,uric acid and adverse reactions during the treatment,the clinical effect was evaluated by observing the improvement of clinical symptoms after 1 week of treatment.Meanwhile,fasting venous blood was collected from three groups.Enzyme linked immunosorbent assay(ELISA)was used to detect Vit C,IL-4and IFN-γ,quantitative real-time polymerase chain reaction(q RT-PCR)was used to detect the expression of nuclear factor erythroid-2 related factor 2(Nrf2)in peripheral venous blood mononuclear cells and P1 protein m RNA of Mycoplasma pneumoniae(MP)in throat swabs,flow cytometry(FCM)was used to detect CD3~+,CD4~+,CD8~+and CD4~+/CD8~+ratio in peripheral venous blood.Results:Before and after treatment,liver function,renal function,uric acid and incidence of adverse reactions were no significant difference in the observation group(P>0.05).In terms of clinical symptom improvement and hospitalization time,the observation group was faster than the control group(P<0.05),In terms of clinical efficacy,after 1 week of treatment,the observation group was better than control group(P<0.05).The levels of Vit C,CD3~+,CD4~+,CD4~+/CD8~+in the healthy group were higher than those in the other two groups before treatment(P<0.05).However,the levels of IL-4,IFN-γ,IL-4/IFN-γand CD8~+in the healthy group were lower than those in the other two groups before treatment(P<0.05).After one week of treatment,the levels of Vit C,CD3~+,CD4~+,CD4~+/CD8~+,Nrf2 in the observation group were higher than those in the control group(P<0.05),IL-4 and the ratio of IL-4/IFN-γwere lower than those in the control group(P<0.05),but the change of IFN-γin the two groups was not significant(P>0.05).Conclusion:(1)Vit C combined with azithromycin can improve the clinical symptoms,shorten the treatment time and is safe in the treatment of MPP in children.(2)Vit C combined with azithromycin treatment of MPP in children may be achieved by regulating cellular immune function and antioxidant stress.
