PurposeColorectal cancer(CRC)is a malignant tumor with high incidence rate and mortality worldwide.Chemokine plays an important role in the development of CRC as one of the important components of tumor microenvironment.CXCL8,also known as interleukin-8(IL-8),is a member of CXC chemokine family.It is mainly produced by macrophages,epithelial cells,airway smooth muscle cells and endothelial cells.CXCL8 and its receptors CXCR1 and CXCR2 play an important role in promoting tumor cell proliferation and metastasis.The purpose of this study was to investigate the correlation between serum CXCL8 expression and different TNM stages of colorectal cancer,and its effect on proliferation,migration and invasion of colorectal cancer.MethodThis study is divided into two parts.The first part is the clinical study of CXCL8 and TNM staging of colorectal cancer patients.60 patients with colorectal cancer and 30 normal healthy people who underwent physical examination in the physical examination center were selected as the control group,The general clinical data of CRC patients and TNM staging of CRC patients were collected.The correlation between serum CXCL8 content and tumor differentiation,staging and metastasis of CRC patients was analyzed.The correlation between serum CXCL8 expression and different TNM staging of colorectal cancer was discussed.The second part is the study of CXCL8 promoting the occurrence and development of colorectal cancer.The expression of CXCL8 and its related receptors CXCR1 and CXCR2 were detected by real time quantitative polymerase chain reaction(q PCR),Meanwhile,SW480 colon cancer cells were selected to inhibit or activate CXCL8 gene.MTT cell proliferation assay and Transwell cell invasion assay were used to explore the mechanism of CXCL8 on SW480 colon cancer cells.Result1.The serum CXCL8 level of colorectal cancer patients was 127.28% ± 30 ng / L,while the serum level of CXCL8 in the control group was 37.29 ± 07 ng / L,t = 29.210,and the difference was statistically significant(P < 0.01),indicating that the serum CXCL8 level of CRC patients was significantly higher than that of normal people.2.The higher the TNM stage,the higher the serum level of CXCL8.At the same time,the study on the correlation between CXCL8 and TNM stage found that CXCL8 was positively correlated with TNM stage of CRC patients,with statistical difference(P < 0.05).The serum CXCL8 level of metastasis group was 141.23% ± 20 ng / L,and the serum CXCL8 level in the non metastasis group was 88.03% ± 38 ng / L,the difference was statistically significant(P < 0.05).3.The content of CXCL8 was 80.29% ± 22 ng / L,and the content of CXCL8 in poorly differentiated patients was 138.33ng/l ± 30 ng / L,the difference was statistically significant(P < 0.05).It can be seen that the lower the degree of tumor differentiation,the higher the content of CXCL8,the worse the prognosis.4.The expression of CXCL8 gene and its receptors CXCR 1 and CXCR 2 in colon cancer tissues were significantly higher than those in adjacent tissues(P < 0.05).When CXCL8 treated SW480 cells for 24 hours,the cell proliferation rate increased significantly(P < 0.05),while when CXCL8 inhibitor was added,the cell proliferation rate of SW480 cells decreased significantly(P < 0.05),indicating that CXCL8 plays an important role in promoting the proliferation of CRC cells.When CXCL8 treated SW480 cells for 24 hours,compared with PBS control group,the number of cell penetration increased significantly(P< 0.05).When CXCL8 inhibitor was added,the number of cell penetration decreased significantly(P < 0.05),indicating that CXCL8 plays an important role in promoting the invasion and migration of CRC cells.ConclusionSerum CXCL8 was significantly increased in CRC patients,and it was correlated with TNM stage of CRC patients.CXCL8 plays an important role in tumor microenvironment by interacting with its receptors CXCR1 and CXCR2,and can promote the proliferation and migration of tumor cells.Therefore,in clinical detection,serum CXCL-8 in patients with CRC is not only helpful for the early diagnosis of CRC,but also helps to predict the severity and prognosis of CRC patients,which has important value in clinical diagnosis.
