| Objective:Sinonasal mucosal malignant melanoma(SMMM)is a rare tumor with a poor prognosis.This study used transcriptome sequencing technology(RNA-Seq)to screen and verify some of the key genes that are differentially expressed in SMMM,and further to explore the prognostic value of tyrosinase related protein 1(TYRP1),ATP-binding cassette subfamily member 5(ABCB5)and matrix metalloproteinase 17(MMP17)in SMMM patients.Method:1、RNA-Seq was used to detecte the expression of m RNA in the mucosal tumor tissues of SMMM patients(n=3)and normal nasal mucosal tissues(n=3)of control patients.Real-time fluorescent quantitative PCR(RT-q PCR)further to verify the expression of key genes,and the target genes were screened out.2、In total,31 paraffin-embedded samples of SMMM treated in the First Affiliated Hospital of Nanchang University from June 2010 to December 2019 were collected.TYRP1,ABCB5 and MMP17 were detected in the tissues of SMMM by immunohistochemistry(IHC).Respectively,the differences in the TYRP1,ABCB5 and MMP17 expressions in the tissues were analyzed and matched with clinical parameters and the total survival rate.Result:1、The differential expression profiles of m RNA in SMMM tissues were detected by RNA-Seq.The results showed that 1537 m RNA expressed more than or equal to 2,accounting for 9.41% of the detectable m RNA.In the experimental group,688 differentially expressed genes were up-regulated and 869 differentially expressed genes were down-regulated.2、Part of the key m RNAs related to the pathogenesis of SMMM were screened through bioinformatics analysis.The expression of 9 selected m RNAs(TYRP1,ABCB5,MMP17,POU3F3,MLIP,CYP19A1,EDNRB,TSPAN10,ADAMTS3)were verified by RT-q PCR,which was consistent with the results of RNA-Seq.3、The overall survival time of 31 patients with SMMM ranged from 6 to 118,with a median survival time of 19 months and an average survival time of 28.9months.The 1-year,3-year and 5-year overall survival were 86.02%,32.53% and21.69%,respectively.Age(p=0.002),typical nasal symptoms(p=0.036)and tumor AJCC stage(p=0.017)might be the synergy prognostic factors of SMMM patients.4、The positive expression rates of TYRP1,ABCB5 and MMP17 in 31 cases of SMMM were 90.32%,80.65% and 64.52%,respectively.The expression of ABCB5 was related with different age groups,and the positive rate of ABCB5 was higher in ≥60 years old group,with a significant difference(p=0.002),and the positive rate of MMP17 expression was higher in non-pigmented group(p=0.02).The positive expression of ABCB5 was negatively correlated with the prognosis of patients with SMMM(p=0.02).Conclusion:There is a significant difference in the expression of m RNA in the mucosal tumor tissues and normal nasal mucosal tissues of SMMM patients.These genes play a key role in the pathogenesis of SMMM.Our results further emphasize a correlation between the expression of TYRP1,ABCB5 and MMP17 in SMMM tissues and point out that a positive ABCB5 expression might be an important factor in judging the prognosis of patients with SMMM. |
