Pancreatic cancer is one of the most malignant tumors. There is no effective treatment in clinic, and the prognosis is very poor.In China, the number of pancreatic cancer patients has been increasing rapidly in recent years. The death rate of pancreatic cancer patients ranks the 6th and 7th in male and female respectively.According to the prediction of international scholars, the death rate related to pancreatic cancer will rise to the second place in 2030,posing a serious threat to human health. It is considered to be a complex genetic disorder. The study of molecular typing of pancreatic cancer will help us to better understand this genetic abnormality. In 2011,Collinson proposed the classification criteria of pancreatic cancer based on the expression profile level, which is divided into three groups, namely "classical subtype","quasi mesenchymal subtype" and "exocrine like subtype".Later,in 2016,Bailey et al also found that the prognosis of the interstitial like subgroup(Bailey et al.Renamed as squamous subtype,i.e.squamous cell pancreatic cancer)was the worst, and it can be used as an independent prognostic indicator to reflect the prognosis of patients. At the gene level, squamous cell carcinoma of the pancreas is characterized by high expression of 20 marker genes. In histology, there are abundant interstitial tissues which show the typical characteristics of hypoxia and lack of blood supply of pancreatic cancer. In biology, they show enhanced glycolysis activity and strong proliferation and invasion ability. We used geo database and big data to analyze pancreatic ductal adenocarcinoma,Ahnak2 was identified by the intersection of the key genes of development and the characteristic genes of squamous cell carcinoma of the pancreas. In addition,we found a gene NFAT5 which may play an important role in pancreatic ductal adenocarcinoma by combining the key genes of glycolysis.Based on the discussion of the biological function of these two genes,we further clarified the squamous cell carcinoma The clinical significance of epithelial tumor type characteristic gene in pancreatic cancer and its role in pancreatic cancer cells. We revealed the biological role of ahnak2 and NFAT5 in pancreatic cancer and their influence on glycolysis, and found the downstream gene PGK1 of NFAT5 in pancreatic cancer. This provides a new idea for the prognosis index and treatment target of PDAC.
