| Objective: AHNAK nuclear protein 2(AHNAK2)is a scaffold protein that regulates cytoskeleton structure,muscle regeneration,calcium channel homeostasis and signal transduction.Recent studies have found that AHNAK2 protein is overexpressed in a variety of malignant tumors,which is involved in the occurrence and development of tumors,and its expression level is closely related to the prognosis of tumors.However,the expression of AHNAK2 protein in gastric cancer and its clinical significance are not very clear.The main purpose of this study is to explore the expression of AHNAK2 protein in gastric cancer and its relationship with clinicopathological features and tumor prognosis.Methods:116 cases of gastric cancer and corresponding paratumorous paraffin specimens were collected from January 1,2016 to December 31,2016.The expression of AHNAK2 was detected by immunohistochemistry(IHC),and the correlation between the expression of AHNAK2 and clinicopathological features(TNM stage,lymph node metastasis,pathological type,degree of differentiation,depth of tumor invasion,distant metastasis,gender,age)and prognosis of patients with gastric cancer was further explored.Results: Among 116 cases of gastric cancer,AHNAK2 protein was positive in 77 cases,the rate was 66.38%(77/116);The positive expression rate of AHNAK2 protein was 2.58%(3/116)in the paracancerous tissue;The expression level of AHNAK2 protein in gastric cancer tissues and adjacent tissues showed significant difference(P=0.000).The correlation between AHNAK2 protein expression and clinicopathological features showed that AHNAK2 protein overexpression was correlated with lymph node metastasis(P = 0.006);AHNAK2 protein level was closely related to the prognosis of patients(log rank test,P = 0.001).Cox regression analysis showed that the expression level of AHNAK2 protein was an independent prognostic factor.Conculusion: 1.AHNAK2 protein is highly expressed in gastric cancer tissues,and the expression level of AHNAK2 protein is correlated with lymph node metastasis of gastric cancer.2.The positive expression of AHNAK2 protein is significantly related to the poor prognosis of patients,which can be used as a prognostic marker and potential treatment target for gastric cancer. |
