Objective: Explore the expression of chemokine receptor CXCR7, E-Cadherin andVimentin in colorectal cancer cells, and by RNA interference targeting silence CXCR7,detect the influence of CXCR7to E-Cadherin and Vimentin in colorectal cancer cell.Methods:1. Useing normal human colon cell lines FHC and colon cancer cell linesHCT-116, LOVO, COLO-205, CACO-2for research. By the real-time PCR technique todetect cells CXCR7, E-Cadherin and Vimentin mRNA expression, at the same time byWestern blot technique to detect cells CXCR7, E-Cadherin and Vimentin, then choose moreinvasive colorectal cancer cell lines into the next phase of the experiment.2. The LOVO andCOLO-205cell lines was transfected with small interfering RNA (siRNA)targeting on CXCR7. Finally using siRNA inhibite CXCR7mRNA, and detecteE-Cadherin, Vimentin expression.Results: Chemokine receptor CXCR7, E-Cadherin and Vimentin were expressed in celllines used in the experiment; CXCR7and Vimentin expression in colorectal cancer cell lineswas significantly higher than in normal colorectal cell lines (P <0.05); E-Cadherin inexpression in colorectal cancer cell lines is lower than normal colorectal cell lines (P <0.05).MRNA and protein expression in colorectal cancer cell lines Vimentin2. CXCR7-siRNAtransfection significantly decreased (P <0.05), while mRNA and protein expression ofE-Cadherin increased (P <0.05). The activation of CXCR7promote E-Cadherin and Vimentinexpression, which may have the function to regulate the expression of E-Cadherin andVimentin in colon tumor cells. When CXCR7was specific inhibit, E-Cadherin and Vimentinexpression become lower.Conclusion: In colorectal cancer cells CXCR7gene highly expressed and silenceCXCR7can increase expression of E-Cadherin, but decrease Vimentin expression. TipCXCR7-siRNA may affect the occurrence and development of colorectal cancer cell lines inthe EMT.
