Objective: The aim of this research was to investigate the role of F-box only protein 9(FBXO9),peroxisome proliferator activated receptor gamma(PPARγ)and nuclear factor-κB(NFκB)in esophageal squamous cell carcinoma(ESCC)and their corresponding paracancerous “normal” tissue,and the relationship between them.Meanwhile,the aim was to explore whether FBXO9,PPARγ and NFκB have clinical significance and may be used as new prognostic markers.Methods: In this research,125 esophageal squamous cell carcinoma paraffin-embedded tissue specimens and corresponding paracancerous “normal” tissue were collected from January 2014 to December 2015.Also,we collect clinical data of selected cases.The immunohistochemical method was used to detect the expression of FBXO9,PPARγ and NFκB in ESCC and their corresponding paracancerous “normal” tissue.The experimental data was analyzed by statistical software SPSS25.0.Results: The positive expression rate of FBXO9 in ESCC was 48.0%,which was higher than that in corresponding paracancerous “normal” tissues(32.0%).The positive expression of PPARγ was 53.6%,which was lower than that in corresponding paracancerous “normal”tissues(68.8%).The positive expression of NFκB in ESCC is 66.4%,which was higher than that in corresponding paracancerous “normal” tissues(57.0%).The expression of FBXO9,PPARγ and NFκB was affected by many clinicopathological factors,such as tumor differentiation,TNM stage and lymph node metastasis.FBXO9 and NFκB were positively correlated with them,while PPARγ was negatively correlated with them.Spearman correlation analysis showed that,in 125 cases of ESCC,FBXO9 was negatively correlated with PPARγ.Moreover,positive correlation between FBXO9 and NFκB,and negative correlation between PPARγ and NFκB was found in our experiment.Univariate survival analysis showed that the overall survival time of patients with positive expression of FBXO9 and NFκB were 27.124±1.710 months and 29.568±1.583 months,respectively.The overall survival time of patients with negative expression of FBXO9 and NFκB were 40.500±1.788 months and 40.762±2.222 months,respectively.(Log-Rank= 21.439,13.535,P<0.001);The overall survival time of PPARγ positive group was 28.538±1.846 months,which was lower than that of negative group(38.515±1.806 months)(Log-Rank =10.451,P< 0.05).Cox multivariate survival analysis showed that the positive expression of FBXO9,PPARγ and NFκB,lymph node metastasis and TNM stage could be independent risk factors for postoperative survival in ESCC patients.Conclusion:1.The abnormal expression of FBXO9,PPARγ and NFκB in ESCC was associated with invasion and development of ESCC.2.The regulation of FBXO9 on ESCC may be related to PPARγ and NFκB.3.The combined detection of FBXO9,PPARγ and NFκB was expected to predict the prognosis of ESCC patients.
