| In recent years,the number of patients with periodontitis suffering from diabetes has been increasing.Studies have shown that there is a close correlation between diabetes and periodontitis.Therefore,finding a drug that can treat diabetes and improve periodontitis has become a research hotspot.Liraglutide has become a clinical drug for the treatment of type 2 diabetes due to its protective function of pancreatic islet β cells.At the same time,the drug has also been shown to have anti-inflammatory and bone-promoting effects.Objective: Exploring the anti-inflammatory effect of liraglutide on diabetic periodontitis and the effect of osteogenic differentiation through animal experiments,and exploring its mechanism,provide a theoretical basis for the clinical treatment of diabetic periodontitis.Methods: We established a diabetic model by feeding rats with high sugar and high fat diet combined with low dose STZ injection.The ligation method was used to ligate the left maxillary first molar of the rat to establish an experimental periodontitis model.After the diabetic periodontitis model was successfully established,Lira was administered for four weeks to detect the changes in rat body weight,fasting blood glucose,and OGTT to explore the effects of liraglutide on the physiological effects of rats.Detect periodontal clinical indicators,the expression of inflammatory factors in rat serum,analyze histopathological staining,and take Micro-CT to explore the effects of liraglutide on periodontal inflammation and bone effects in rats.The mechanism of liraglutide in the treatment of diabetic periodontitis was studied by q RT-PCR and Western blot experiments.Results:(1)The diabetic periodontitis model was successfully set up by feeding high sugar and fat feed combined with 35 mg/kg STZ injection and periodontal ligation.Successfully modeled rats showed typical clinical symptoms of diabetes and periodontitis,fasting blood sugar Increased glucose tolerance,periodontal tissue destruction,and alveolar bone resorption.(2)The weight of rats was reduced after liraglutide treatment,the fasting blood glucose of rats in the diabetic periodontitis group decreased,and the abnormal glucose tolerance was effectively improved.(3)The administration of liraglutide alleviated the clinical symptoms of periodontitis in periodontitis rats and diabetic periodontitis rats,inhibited the inflammation of periodontal tissue in periodontitis rats and diabetic periodontitis rats,and reduced the expression of inflammatory factors in periodontitis and diabetic periodontitis,and reduce the destruction of alveolar bone on the ligation side of periodontitis and diabetic periodontitis.(4)Liraglutide administration down-regulated the JAK2/STAT3 gene and protein expression levels in rat gingival tissues.Conclusion: Liraglutide improves the symptoms of diabetes and periodontitis in rats;it may affect the occurrence and development of diabetic periodontitis through the JAK2/STAT3 signaling pathway. |
