| The genus Ervatamia(synonym: Tabernaemontana)(Apocynaceae)includes about 120 species,and widely distributes in the tropical and subtropical areas of Asia and Australia.The main secondary metabolites of this genus are indole alkaloids and terpenes,which have biological activities such as antibacterial,anti-inflammatory,anti-fertility,anti-oxidation,analgesic,anti-tumor and enzyme inhibition.In order to further discover compounds with novel structures and better biological activities from the genus Ervatamia,this thesis studied the chemical constituents of Ervatamia chinensis(Merr.)Tsiang,a medicinal plant from Yunnan.After extracting the branches and leaves of Ervatamia chinensis with methanol,it was acid-dissolved and alkali-precipitated and separated by normal phase silica gel column chromatography,RP-18 and Sephadex LH-20 column chromatography.Finally,the structure of the compounds was identified by using a variety of modern spectroscopy techniques.As a result,32 compounds,including 28 alkaloids and 4non-alkaloids,were isolated from the chloroform part and ethyl acetate part of the plant.And their structures were elucidated as voastrictine N4-oxide(1),voastrictine(2),coronaridine hydroxyindolenine(3),voacangine hydroxyindolenine(4),hainanervatacine(5),voacangine(6),coronaridine(7),voacristine(8),heynaenine(9),3-(2-oxopropyl)voacangine(10),3-(2-oxopropyl)coronaridine(11),ibogamine(12),hainanervatasine(13),(-)-mehranine(14),voalenine(15),N1-methylvoaphylline(16),N1-methylvoafinine(17),ervayunine(18),ervatamine(19),20-epi-ervatamine(20),pseudoindoxyl derivative(21),tabernaemontanine(22),janetine(23),vallesamine(24),appricine(25),ochrobonine A(26),tubotaiwine(27),conofoline(28),α-amyrin acetate(29),cycloeucalenol(30),4-hydroxy-3-methoxybenzaldehyde(31)and acetovanillon(32)respectively.Among them,compound 1 is a new quinoline alkaloid,and the activity screening of compound 1 and compound 2 with similar structure showed that neither compound 1nor compound 2 inhibited the growth of Phytophthora infestans 88069 strain.Compounds 26 and 32 were isolated from genus Ervatamia for the first time.Compounds 2-6,8,11-15 and 17-32 were obtained from the Ervatamia chinensis originally time.At last,the research progress of the alkaloids and their biological activities in domestic genus Ervatamia plants were reviewed. |
