ObiectiveAs the world enters an accelerated aging society today,brain aging and the decline in aging-related cognitive and motor abilities have increased the burden on society and families,and also caused many inconveniences for the elderly and affected the quality of life.Cognitive decline in Alzheimer’s disease(AD)as the main representative is still lacking effective treatments.Mild cognitive impairment(MCI),as the precursor of AD,is reversible and is one of the stages to prevent progressive decline of cognitive function.Therefore,it is of great significance to prevent and monitor the occurrence and development of neurodegenerative diseases such as MCI.In this paper,a case-control study was conducted to explore the relationship between physical activity and cognitive function in elderly MCI patients,and whether to use grip strength,pace,and stand-gait test as diagnostic indicators to predict cognitive decline.Based on the information provided by the population experiment,select the rapidly aging mice(senescence-accelerated mouse prone 8,SAMP8)with the characteristics of cognitive dysfunction for 6 months to record the enhanced changes in muscle function during the intervention period to explore whether the SAMP-8 mouse model can be used as a feasible animal model for further research on the prevention and treatment of elderly-related diseases,screening of therapeutic drugs and research on related exercise mechanisms.Methods1.This study is a case-control study.The study subjects were all from the 60 to 80-year-old physical examination population in Baodi District,Tianjin City from March 2019 to April 2019.Historical questionnaire surveys,assessment of cognitive function,and testing of motor function will take older people who can provide complete and objective information as interviewees.After the baseline survey was completed,120 MCI patients were screened into the MCI group based on the prevalence of MCI and their independent risk factors.Among the respondents included in the same period,120 cognitively healthy persons were matched according to age and gender as 1: 1 Control group.The mini-mental state examination(MMSE)was used to evaluate the cognitive function of MCI patients.The personal history questionnaire was used to collect the sociodemographic information and disease history of the elderly.The activity of daily living scale was used.Daily living(ADL)to assess the elderly’s ability to perform daily activities;use the International Physical Activity Questionnaire(IPAQ)short questionnaire to evaluate physical activity;collect three indicators of grip strength,walking speed,and standing-walking test to reflect the elderly Human body muscle strength,ability to move and dynamic balance.Using the above methods to collect the living status of the elderly in Tianjin and evaluate the elderly’s athletic ability can be used as auxiliary diagnostic indicators for cognitive screening.2.Animal experiments: 4 months old SAMP8 mice and 4 months oldsenescence-accelerated mouse resistant(SAMR1)were raised separately.The normal feeding time is 6 months.When the mice reach 10 months old,twenty male mice were divided into two groups.Ten 4-month-old male SAMP8 mice were used as the young control group(P8-4),and behavioral tests were conducted.The 10-month-old SAMP8 mice(P8-10)were compared with the 10-month-old SAMR1 mice(R1-10)and the 4-month-old SAMP8 mice,respectively.The degree of aging of the mice was evaluated by the aging score,and the number of red blood cells and hemoglobin of the mice were recorded.Content,coefficient of variation of red blood cell distribution width,and standard deviation were used to evaluate red blood cell function,and relative muscle mass,grasping force,rotating rod fatigue test,and swimming test were used to evaluate muscle function in mice;this study used small(10-month-old)SAMP8 and SAMR1 Rats and young(4-month-old)SAMP8 were studied in order to provide valuable reference information for the study of changes in aging-related functions.ResultsThe results of the case-control study showed that(1)the education level,income level,and number of household residents of the MCI patients are lower than those of the control group(P <0.05).In terms of physical activity,the grip strength,gait speed,and physical activity of the MCI group And number of high-intensity exercises per week is lower than the control group(P <0.05),the timed up and go test took longer than the control group(P <0.05),and the hemoglobin content and number of red blood cells in the MCI group were lower than those in the control group(P <0.05).(2)Educational level(95% CI: 0.100 ~ 0.490,0.078 ~ 0.524,0.024 ~ 0.202),income level(95% CI: 0.109 ~ 0.464),number of permanent households(95% CI: 0.133 ~ 0.567,0.117 ~ 0.666)timed up and go test(95% CI: 1.372 ~ 2.055),gait speed(95% CI: 0.948 ~ 0.979),grip strength(95% CI: 0.805 ~ 0.905),exercise intensity(95% CI: 0.286 ~ 0.833),Hemoglobin content(95% CI: 0.938 ~ 0.987)was correlated with cognitive function.(3)Grip strength(OR: 0.673,95% CI: 0.518 ~ 0.873,P <0.05)and gait speed(OR: 0.954,95% CI: 0.923 ~ 0.986,P <0.05)are protective factors of cognitive function,grip strength The longer and the faster the gait speed,the less susceptible to MCI;the length of time for the timed up and go test(OR: 1.671,95% CI: 1.208 ~ 2.311,P <0.05)is a risk factor for cognitive function,and the longer the time,the easier it is Suffering from MCI.(4)The predictive effects of the joint diagnosis of MCI(ROC area: 0.689,0.725,0.841,0.842)of the three items of grip strength,gait speed,and timed up and go test are better than the individual prediction results.(5)There was a positive correlation between grip strength,gait speed,and hemoglobin content in the MCI group(r = 0.328,P <0.05;r = 0.227,P <0.05).The time spent in the timed up and go test was negatively correlated with the hemoglobin content(r =-0.271,P <0.05),gait speed was positively correlated with the number of red blood cells(r = 0.042,P <0.05);grip strength in the control group was positively correlated with hemoglobin content(r = 0.459,P <0.05),and positively correlated with the number of red blood cells(r = 0.241,P <0.05).The results of animal experiments showed that:(1)the score of aging scores showed that the scores of SAMP8 mice at 10 months of age were higher than that of SAMR1 mice at the same month(P <0.05),showing a more severe aging state;(2)the results of red blood cell function showed 10 Compared with 4-month-old SAMP8,the number of red blood cells and hemoglobin content of SAMP8 were decreased(P <0.05),and the width of red blood cell distribution was increased(P <0.05).At 4 months of age,SAMP8(P <0.05),muscle strength and muscle endurance decreased(P <0.05),swimming speed slowed(P <0.05);(4)correlation analysis results showed that the number of red blood cells,hemoglobin content and grip There was a positive correlation(r = 0.407,r = 0.416,P <0.05).ConclusionCase-control studies have shown that greater grip strength,faster gait speed,and shorter timed up and go test time are MCI protective factors.Hemoglobin in MCI patients is positively correlated with grip strength and gait speed,and timed up and go test time is negatively correlated.SAMP8 mice show a decline in aging red blood cells and muscle strength during aging,which is consistent with human natural aging characteristics,and can be used as a predictive indicator to judge cognitive decline.It is used to study age-related diseases,such as hematopoiesis in mild cognitive impairment And provide information on muscle function decline.
