Taxol is a kind of terpene compounds, which was separated from Taxus brevifolia originally. Becauseof the special anticancer mechanicsand special curative effect for some kinds of tumor, Taxol attracts muchattention recently. It is very difficult to separate and purify Taxol from Taxus, because the content of theTaxol in Yaxus is very low, and there are also some analogues of Taxol in Taxus, which make it morediffeult to separate.Consequently, it is very important to develop a method to separate and purify Taacolwith high recoveries.This thesis includes five parts.The first part includes the review of the latest researches on Taxus, the studies on the extraction andpurification of taxol; the studies on the anti-tumor effect of taxol; the determination of residual volume ofthe organic solvent in the drug and the problems met in the manufacture procedures, and 113 referenceswere cited.The second part is about studies on the extraction technology of Four Tank Counter Current to extracttaxanes from Taxus. We extracted taxol and analogs from Taxus with the method of Four Tank CounterCurrent, and investigated in detail the factors which may affect the technology, including methanolconcentration, the contact time, the temperature and so on. And then we determined the best condition andestablished a new extraction technology of taxol.In the third part RP-HPLC was used to determine the contents of taxol in the extract of Taxus, and amethod used to determine the contents of taxol in the extract of T. Canadensis Leave was established, whilethe results showed that this method was easy and accurate and had good precision. Then this method wasadopted to determine the contents of taxol in the extracts of different kinds of Taxus.In the fourth part HPLC and RP-HPLC was combined to establish a new method of extracting taxol,cephalomanirme, 10-DAB and 7-epitaxol from the Taxus extract. A new homemade porous polymer resinfilling (JTY-1) was applied as stationary phase in the RP-HPLC. The results of the experiment showed thatJTY-1 showed high selectivity towards taxol and cephalomaninne, and had small irreversible adsorption,and what's more, this kind of filling is reclaimable.The last part is about silica gel regeneration. After regeneration, the silica gel used in taxol industrycompare the new gel regeneration in adsorbability,separating property is not bad than the new silica, showthat device of silica gel regeneration is useful.This article innovates the spot:1,Has established one kind of new taxus mellow extraction method-four section of adverse currentlaw, and has carried on the craft inspection to this method, indicated this method is suitable in the T.Canadensis marsh taxus mellow extraction, has provided one new way for the industrialization production.2,Established one kind of RP-HPLC to determine in the T.
