Separation and extraction of active monomeric compounds from medicinal plants has always been an important research direction.However,many studies have shown that the compounds in the plant do not exist or work alone,but will show a certain proportion and interact with each other.Traditional Chinese medicine(Solanum lyratum Thunb.)has a good effect on the anti-tumor effect in clinic,however,the cytotoxicity of the monomer compounds isolated from it in the previous studies was weaker compared with the whole plant effect,we speculate that there may be some compounds with synergistic effects,the following experiments were carried out.(1)Alkaloids,total saponins and the whole plant extract were separated from Solanum lyratum Thunb.and the MTT assay was used to determine the toxic effects of these components on HMEC-1 cells.It was found that the whole plant extract of Solanum lyratum Thunb.at the same concentration is less toxic than others,indicating that the whole plant extract can reduce cytotoxicity and protect cells.(2)We separated four different types of compounds from Solanum lyratum Thunb.by silica gel chromatography,sephadex LH-20 and thin layer chromatography,namely solaretivone(SO),n-dodecanol(DO),tigogenin(TI)and friedelin(FR),and identified their structure by NMR and MS,among them SO and FR were first separated from it.The MTT assay was used to detect the in vitro anti-tumor activity of SO,TI and FR on A549 and Hep G2 cells when used alone or in combination.The results showed that when the three compounds were used alone,only TI had an insignificant inhibitory effect on Hep G2 at high concentrations;it had a synergistic effect when any two used in combination;the three kinds of combination has the best anti-tumor effect,which inhibition rate can over 40%.(3)In the preliminary work of the laboratory,a good anti-inflammatory monomer compound Diosgenin3-O-α-L-rhamnopyranosyl-(1→2)-β-D-glucopyranosiduronic acid(DRG)was isolated from Solanum lyratum Thunb.In this study,the HPLC method was used to determine the content of the compound in the total saponins,the anti-inflammatory comparison between the total saponins and the compound was carried out with the established anti-inflammatory model before.As the results showed that when the content of the compound DRG remained the same,the anti-inflammatory effects of the two were not equal,but the anti-inflammatory effect of total saponins is weaker than DRG alone.In addition,we measured the anti-tumor activity of DRG alone and in combination with SO,FR,and TI.The results showed that TI has the most significant inhibition effect on DRG,and there is no interaction between FR and DRG.These results demonstrate that there are antagonism effect between the chemical constitutes of Solanum lyratum Thunb.besides synergistic effect.
