Study On The Extraction And Inclusion Technology And Stability Of Volatile Oil From Qizhi Weitong Granules

Qizhi Weitong granules is listed in the 2015 edition of the《Pharmacopoeia of the People’s Republic of China》(hereinafter referred to as the《Chinese Pharmacopoeia》),which is composed of Bupleurt Radix,Aurantii Fructus,Paeoniae Radix Alba,Glycyrrhiae Radix Et Rhizoma Praeparata Cum Melle,Cyperi Rhizoma,and Corydalis Rhizoma.It is clinically used to liver stagnation,chest swelling,and epigastric pain.Aurantii Fructus and Cyperi Rhizoma are rich in volatile components,which are one of the main therapeutic components of Qizhi Weitong granules,and affect the overall quality and subsequent clinical efficacy of the preparation.The volatile oil is unstable,and the effective extraction methods and preparation technology are the guarantee of the content and clinical efficacy of the volatile oil.The study on the extraction,inclusion and stability of the volatile oil in Qizhi Weitong granules is of great significance for improving the drug quality ang further development and application.Objective: To optimize the extraction process,inclusion process of total volatile oil from Aurantii Fructus and Cyperi Rhizoma in Qizhi Weitong granules,the aim is to improve the content and stability of volatile oil in preparation and enhance the efficacy of volatile oil.Methods: On the basis of single factor experiment,in order to total volatile oil yield,and total volatile oil effective components limonene,α-Cyperone,Cyperene for the target,design of response surface method(RSM)combined with entropy weight method,screening of the WER best extraction technology of total volatile oil.On the basis of single factor test,RSM combined with entropy weight methods was used to select the optimal inclusion process parameters of the total volatile oil.The stability of qizhi weitong granules which contain volatile oil inclusion compounds and which was direct injection of volatile oil were investigated,by compare the content changes of limonene,α-Cyperone,Cyperene.Results: The optimal extraction process is WER,the ratio of liquid to material is 13:1(m L/g),the soaking time is 1.1h,and the extraction time is6.5h.The optimal inclusion process is: Grinding method,the ratio of β-CD to volatile oil is 7.5:1(g/g),the ratio of liquid to volatile oil is 5:1(m L/g),grinding time is 1h.Preparation after the inclusion of volatile oil,the effective component content is relatively stable under test conditions,while the volatile oil without inclusion,the effective component content was continuous decline or even zero under test conditions.Conclusions: The optimal extraction technology of WER can effectively improve the extraction rate and effective component content of total volatile oil of Aurantii Fructus and Cyperi Rhizoma in Qizhi Weitong granules.The optimal screening method and process can effectively inclusion the total volatile oil,and improve the stability of the total volatile oil of Aurantii Fructus and Cyperi Rhizoma in Qizhi Weitong granules.
