| Objective:Study on the effect of Buyang Huanwu Decoction Combined with neural stem cell transplantation on cerebral ischemia/reperfusion injury in rats based on Notch signaling pathway.Methods:Neural stem cells were cultured and identified from the cerebral cortex of 14 day pregnant rats.80 clean male SD rats were randomly divided into 5 groups,the sham operation group,the model group,the Buyang Huanwu Decoction group,the transplantation group,the Buyang Huanwu Decoction transplantation group.Every group has 16 rats respectively.In the sham operation group,the rats’seperated blood vessels was inserted by thread plug.The other groups were used to establish the complete model of cerebral ischemia/reperfusion by the improved thread occlusion method.The Buyang Huanwu Decoction group and The Buyang Huanwu Decoction transplantation group were given Buyang Huanwu Decoction by gavage after awake operation,the dosage was 14.8g·kg-1·D-1.Neural stem cells were transplanted into the striatum of ischemic side 24 hours after model establishment.After 14days,the neurological function of rats in each group was observed by Zea longa neuro behavioral score,the volume of cerebral infarction and the damage of brain cells were monitored through test mode of TTC and HE staining,while the changes of Notch1,Hes1 and Hes5 in ischemic side were detected by Western blot.Results:In the sham operation group,there was no defect of nerve function,no cerebral infarction and normal brain cells.Compared with the sham operation group,the nerve function of the model group was obviously damaged,the volume of cerebral infarction was obviously increased,the brain tissue cells were seriously damaged,the expression of Notch1,Hes1,Hes5 in the brain was increased(P<0.05);Compared with the model group,the rats in the transplantation group,the rats in the Buyang Huanwu Decoction group and the rats in the Buyang Huanwu Decoction combined with neural stem cell transplantation group were relieved in different degrees,the expression of Notch1,Hes1,Hes5 in the brains was increased(P<0.05);Compared with the transplantation group and the Buyang Huanwu Decoction group,the neurological score of the Buyang Huanwu Decoction combined with neural stem cell transplantation group was significantly decreased,the volume of cerebral infarction was reduced,the damage of brain tissue cells was significantly alleviated,and the proteins’level were significantly increased(P<0.05).Conclusion:The protective effect of Buyang Huanwu Decoction Combined with neural stem cell transplantation is better than Buyang Huanwu decoction or neural stem cell transplantation alone on brain injury in rats with cerebral ischemia/reperfusion,and its mechanism may be related to the up regulation of Notch1,Hes1 and Hes5 expression. |
