Objective:To explore the expression of inflammatory factor Interleukin-6(IL-6)in the tumor tissues of patients with hypopharyngeal squamous cell carcinoma,as well as the effect and mechanism of interleukin-6 on the proliferation and chemotherapy resistance of hypopharyngeal squamous cell carcinoma.In order to provide a possible direction and reference basis for the treatment of clinical hypopharyngeal squamous cell carcinoma.Methods:Collect the paraffin pathological sections of patients with hypopharyngeal squamous cell carcinoma,and detect the expression of IL-6 in 44 cases of hypopharyngeal squamous cell carcinoma tumor tissues by immunohistochemistry;Then by CCK8,MTT,Western blot and other methods,the proliferation,drug resistance,and the expression of Stat3 and P65 protein in hypopharyngeal squamous cell carcinoma cells pretreated with IL-6(PreIL-6 Fadu)and hypopharyngeal squamous cell carcinoma cells(Fadu)were detected in vitro experiments.Results:The expression of IL-6 in tumor tissue is related to whether hypopharyngeal squamous cell carcinoma is accompanied by necrosis,degree of differentiation,and lymph node metastasis(P<0.05),and has no significant correlation with the patient’s gender,age,and TNM grade(P>0.05),the expressionof IL-6 in the tumor tissues of poorly differentiated hypopharyngeal squamous cell carcinoma is higher than that in the tumor tissues of middle and high differentiation,and the expression of IL-6 in the tumor tissues of hypopharyngeal squamous cell carcinoma patients with lymph node metastasis is higher than that in the tumor tissues without lymph node metastasis.The expression of IL-6 in tumor tissues with necrosis is higher than that in tumor tissues without necrosis.In in vitro experiments,after 72 hours of culturing Fadu cells with IL-6 with a concentration gradient of Ong/ml to 50ng/ml,the overall mean OD values of Fadu cells in the six groups were statistically different(F=15.1,P<0.05).Fadu cells were cultured in Ong/ml and 50ng/ml IL-6 medium,and the proliferation of fadu cells was detected at different time points.There was no significant difference in OD values between the two groups at 0h,24h,48h,and 96h(P>0.05).At 72h,the OD value of Fadu cells pretreated with 50ng/ml IL-6 was significantly higher than that in the Ong/ml group,the difference was statistically significant(P<0.05),indicating that IL-6 can promote the proliferation of Fadu cells,and 50ng/ml IL-6 pretreated Fadu After 72 hours,IL-6 had the most obvious proliferation effect on Fadu cells.The IC50 of preIL-6Fadu cells to cisplatin,fluorouracil,and doxorubicin was higher than that of normal Fadu cells,and the difference in IC50 between the two cells was statistically significant(P<0.05).The expression of stat3 and P65 in preIL-6Fadu cells was increased compared with Fadu cells,and the difference was statistically significant(P<0.05).Conclusion:IL-6 participates in the occurrence and development of hypopharyngeal squamous cell carcinoma,and promotes the proliferation and drug resistance of hypopharyngeal squamous cell carcinoma through Stat3 and NF-κB related pathways.IL-6 is a possible target for the treatment of hypopharyngeal squamous cell carcinoma,and its in-depth study has high research value for the decision-making of the treatment of hypopharyngeal squamous cell carcinoma chemotherapy resistance in clinical work.
