Objective:To explore the effect of PRMT2 on autophagy and apoptosis of umbilical vein endothelial cellsMaterials and methods:HUEVCs(Human umbilical vein endothelial cells)were used as the research object.The experiment was divided into PRMT2 high expression group,PRMT2 empty vector group,and blank control group(no special treatment);liposome transfection was used to complete the transient Transfection,use immunofluorescence staining to determine the expression and localization of PRMT2 and LC3 in umbilical vein endothelial cells(HUEVCs);use transmission electron microscope to observe the formation of autophagic vesicles in each group of cells;use Western Blot to detect autophagy in each group of cells Protein expression;flow cytometry was used to detect cell apoptosis in each group.Results:1.Immunofluorescence results show that PRMT2(red)and LC3 are expressed in the cytoplasm of cells in each group,and in the PRMT2 high expression group,LC3 punctate aggregation and green spots with enhanced fluorescence;2.Transmission electron microscope observation of autophagic vesicles It can be seen that the autophagy vesicles in the PRMT2 high expression group were significantly increased compared with the blank control group and the empty vector group;3.Western Blot results showed that the autophagy-related proteins LC3,Beclin-1 and p62 in the PRMT2 overexpression group were significantly increased compared with the empty vector group.It is statistically significant(P<0.01);4.Flow cytometry detection 48 h after cell transfection,the total apoptosis rate of the PRMT2 high expression group is(4.07 ± 0.02517)%,which is significantly lower than that of the empty vector group(18.24 ± 0.1345)% and the blank control group(12.21 ±0.1908)%,the difference was statistically significant(P<0.01).After 48 hours of cell transfection,PRMT2 detected by MTT had no significant effect on the proliferation of HUEVCs,and the difference was not statistically significant(P>0.05)Conclusion:High expression of PRMT2 inhibits apoptosis of HUEVCs and promotes autophagy of HUEVCs...
