Objective: Through GEO database chip mining and circular RNA database,circular RNAs related to papillary thyroid carcinoma and oral squamous cell carcinoma were screened out.This study will explore the relationship between hsa_circ_0011385 and clinicopathological events in patients with oral squamous cell carcinoma or papillary thyroid carcinoma,as well as the clinical diagnostic value of hsa_circ_0011385 in oral squamous cell carcinoma and papillary thyroid carcinoma.Methods: We retrieved the chip data of papillary thyroid carcinoma tissue and circular RNA from GEO database,use GEO2 R online analysis tool to analyze online,and combined the circular RNA database and literature reading to screen out the differentially expressed circular RNA.47 pairs of cancer and para-cancerous tissues were collected from patients who were diagnosed with oral squamous cell carcinoma in the first People’s Hospital of Xiangtan City from October2018 to November 2020.The relative expression of hsa_circ_0011385 in oral squamous cell carcinoma tissues and para-cancerous tissues was detected by uing real-time fluorescent quantitative PCR.Then analyze and compare whether there are significant differences between the relative expression of hsa_circ_0011385 in oral squamous cell carcinoma tissues and para-cancerous tissues.Analyze the relationship between the expression of hsa_circ_0011385 in oral squamous cell carcinoma tissues and related clinicopathological events in patients with oral squamous cell carcinoma.Receiver operating characteristic curve(ROC curve)was used to explore the diagnostic value of hsa_circ_0011385 for oral squamous cell carcinoma.The method of treating 49 patients with papillary thyroid carcinoma is the same as aboveResults: 1.The expression of hsa_circ_0011385 in oral squamous cell carcinoma tissues and papillary thyroid carcinoma tissues was significantly higher than that in adjacent tissues(P <0.01).2.The expression level of hsa_circ_0011385 is related to lymph node metastasis and TNM staging in patients with oral squamous cell carcinoma(P <0.05),but it is not significantly related to gender,age,drinking,smoking,chewing betel nut,tumor size,and tumor location(P >0.05).The expression level of hsa_circ_0011385 is related to lymph node metastasis in patients with papillary thyroid carcinoma(P<0.05),but it is not significantly related to gender,age,tumor size,TNM staging,number of lesions and tumor location(P >0.05).3.The ROC curve results show that the area under the curve(AUC)of hsa_circ_0011385 in oral squamous cell carcinoma tissue is 0.772(95% CI: 0.676 to 0.868,P<0.01).When the Youden index is set to1.3264,the corresponding sensitivity and specificity are 0.617 and 0.872 respectively.The area under the curve(AUC)of hsa_circ_0011385 in papillary thyroid carcinoma tissue is 0.835(95%CI: 0.754~0.916,P<0.01).When the Youden index is set to 1.3467,the corresponding sensitivity and specificity are 0.857 and 0.714 respectively.Conclusion: 1.The expression level of hsa_circ_0011385 is related to lymph node metastasis and TNM staging in patients with oral squamous cell carcinoma and related to lymph node metastasis in patients with papillary thyroid carcinoma.
