At present,in the context of tobacco and health,it is of great significance to develop chemical and instrumental analysis techniques for tobacco quality and harmful metabolic components produced by smoking.In the early stage of this project,the instrumental analysis methods of metabolites related to tobacco quality have been studied,and the research on the analysis methods of harmful metabolites produced by tobacco smoking will be focused on in this thesis.In order to scientifically study the metabolic components related to the harmfulness of tobacco smoking,this thesis takes the blood of different smokers as the research object and establishes four types of chemical components benzo[a]pyrene and its metabolites,tobacco-specific nitrosamines and its metabolites,The detection method of nicotine and its metabolites and key metabolites of oxidative stress response is to explore the relationship between various targets by measuring the contents of various chemical substances and metabolites in the blood of different smokers and combining mathematical statistics.The metabolism of various chemical components in the body and smoking exposure provides a reference for the correlation between tobacco and health.Objective:Taking the blood of different smokers as the research object,a chemical instrumental analysis method was established to detect the contents of several types of tobacco chemical components and their key metabolites in the body,and to explore the metabolic differences of various chemical components of the human body by smoking exposure.Methods:(1)Establ ished an online liquid-liquid two-dimensional chromatography method for benzo[a]pyrene and its metabolites in the blood;an online liquid-liquid two-dimensional chromatography method for tobacco-specific nitrosamines and its metabolites in the blood;On-line liquid phase-liquid two-dimensional chromatography for key metabolites of oxidative stress in blood;liquid chromatography-tandem mass spectrometry for nicotine and its metabolites in blood.(2)A one-way analysis of variance was used to explore the metabolic differences of the four types of chemical components caused by smoking exposure.(3)Use canonical correlation analysis to explore the relationship between the four types of chemical components.(4)Adopt multivariate statistical principal component analysis and cluster analysis to comprehensively evaluate the similarity of the four chemical components affected by smoking exposure.Results:(1)Through the establishment of analysis methods for several types of chemical components in the blood of different smokers.The results show that the on-line liquid-liquid two-dimensional chromatography detection method with sample purification system and solid-phase extraction enrichment function can meet the determination of trace and trace tobacco chemical components and their metabolites in the blood of smokers.It can realize sample purification,solid phase extraction and instrument analysis and detection at the same time,which greatly simplifies sample pretreatment steps.The constructed method has high detection sensitivity,good precision,strong selectivity and ideal experimental results.(2)One-way analysis of variance shows that smoking exposure has significant metabolic differences on several types of chemical components in the blood of different people,and the metabolic differences of tobacco-specific nitrosamines,nicotines,and oxidative stress reactions have obvious laws:severe Smoking group>moderate smoking group>light smoking group>non-smoking group;and the difference in metabolism of benzopyrenes is only statistically significant in the heavy smoking group.(3)Canonical correlation analysis shows that several types of chemical components have significant correlations with each other.Nicotines are strongly related to nitrosamines and oxidative stress,and moderately related to benzopyrenes;oxidative stress is moderately related to nitrosamines and benzopyrenes;nitrosamines and benzopyrenes Weak correlation.(4)Multivariate statistical analysis showed that the heavy smoking group had the highest comprehensive scores of the four types of chemical component indicators,which was significantly different from the comprehensive scores of the non-smoking group and the light smoking group,but its content was still at the nanogram level,and it was in-depth research on health.Need to be further explored.Conclusion:On-line liquid-liquid two-dimensional chromatography with purification system and solid-phase extraction function is suitable for the determination of trace and micro-tobacco chemical components in complex matrix blood,and blood sample purification,solid-phase extraction and enrichment,instrument analysis and detection It can be carried out at the same time,which greatly simplifies the sample preparation steps.The method has high detection sensitivity,good precision,strong selectivity and ideal experimental results.Mathematical statistics show that the metabolism of various chemical components in the body is significantly affected by smoking exposure and has similar laws,and there is a significant correlation between various chemical components,and various indicators interact and influence each other in the body.
