| Objective:1.To investigate the effects of the three-component combination of the Chinese herbal medicine Radix et Rhizoma Pseudostellariae,Radix Astragali and Radix Scrophulariae on myocardial mitochondrial respiratory chain complex I activity as well as superoxide dismutase(SOD)and malondialdehyde(MDA)in heart failure rats,in order to investigate the mechanism of their treatment of chronic heart failure.2.We searched the TCM or combined Chinese and Western medicine literature related to heart-kidney-yang deficiency evidence of chronic heart failure,screened the entry pool of the heart-kidney-yang deficiency evidence of heart failure disease diagnostic scale,and performed reliability and validity tests to provide a theoretical basis for quantifying and standardizing the diagnosis of heart-kidney-yang deficiency evidence of heart failure disease in the future.Methods:1.60 clean SD rats were divided into control group,model group,Astragaloside(ATL),Trimetazidine(TMZ)and Traditional Chinese medicine under high dose(TCMH)and Traditional Chinese medicine under low dose(TCML)by random number method.(TCMH)and Traditional Chinese medicine under low dose(TCML),10 rats in each group were used to establish the chronic heart failure rat model with pharmacological interventions using the adriamycin induction method.(1)The left ventricular end-diastolic diameter(LVDd),left ventricular end-systolic diameter(LVDs),ejection fraction(LVEF)and left ventricular short-axis shortening rate(LVFS)were measured in plasma BNP samples and echocardiograms at week 4.((2)The changes of myocardial mitochondrial respiratory chain complex I activity were observed by enzyme-linked immunosorbent assay(ELISA).(3)The superoxide dismutase(SOD)and thiobarbituric acid(TBA)method of myocardial tissue were used to determine the content of malondialdehyde(MDA)by WST-8 method.2.Systematic search of China Knowledge Network(CNKI),Wanfang and Wipu databases from 1998-2019 with literature studies on TCM and combined Chinese and Western medicine in heart failure,collation of literature and import of literature titles using Note Express standard version(version 3.3.0.8102)software,elimination of duplicate literature with the help of software,and then manual preview of titles,reading of reviews and We then manually previewed the titles,read the review and full text,and sorted the literature strictly according to the inclusion and exclusion criteria,and finally collated a total of 183 papers.The pool of entries was initially screened using an expert questionnaire,and then the pool of scale entries was fine-selected using multivariate statistical methods,and the reliability and validity of the scale were examined.Conclusions: 1.Compared with the blank group,BNP and cardiac function indexes of rats in the model group,trimetazidine group,astragaloside group,herbal high-dose group and herbal low-dose group were reduced,and cardiac function was the lowest in the model group.After successful modeling of heart failure rats,the mitochondrial complex I enzyme activity and SOD activity in each group of drug intervention were obvious and the MDA content decreased compared with the model group,and the effect of the low-dose group of Chinese medicine was significant.2.A diagnostic scale of heart-kidney yang deficiency in heart failure disease including a total of 11 entry pools was developed,including heart-yang deficiency dimension: palpitation,shortness of breath,fatigue;kidney-yang deficiency dimension: fear of cold extremities,swelling,and little urination;tongue and pulse signs dimension: pale face,fat tongue,pale tongue,white coating,and weak pulse,with satisfactory reliability and validity. |
