Objective:To look up for the mechanism of Moisturizing and Turbidity Reduction in the treatment of HU A use network pharmacology,to clarify the biological function and signal pathway of dehumidification and turbidimetry,to screen out functional targets and to verify them by animal experiments.To provide some objective basis for clinical application of this prescription.Methods:Use TCMSP database to get the effective chemical components and to find out the targets for the treatment of each effective chemical component,and the targets were standardized with the help of uniprot database.Then use Cytoscape3.7.1 to construct the visual network of component-target.The target genes of hyperuricemia were searched by OMIM、Disgenet、Drugbank database,and the intersection targets of humidification and turbidification prescription and HU A were screened.protein interaction networks were looked up by STRING database.We can also use Metascape databases to get the gene ontology function enrichment and KEGG pathway analysis.HUA rat model was established to avoid the expression of some targets by blotting(Western Blot)and Real Time qPCR.Results:The results showed that there were 61 active components,which were predicted to act on 258 targets and 37 targets with hyperuricemia,including 11 core targets and 20 signaling pathways related to hyperuricemia,such as inflammation,immunity,proliferation and apoptosis.The results of the rat experiment showed that the six groups of experimental data after 1 week found that the uric acid value was significantly higher than that compared with the blank controls(P<0.05),indicating the successful manufacture of the model of hyperuricemia rats.On comparison of blank group,the uric acid of the model group were increased very high(P<0.01).In each dose group of traditional Chinese medicine and Benzbromarone treatment,the serum uric acid,serum creatinine and blood urea nitrogen were decreased very high compared with the model group(P<0.05),which indicated that the wet and turbid prescription had the effect of reducing uric acid and had low damage to kidney.The protein expression and mRNA of key target ABCG2 were significantly lower in the model group than in the blank group(P<0.05),while the expression and mRNA relative expression of the key target protein in the benzbromarone group and Moisturizing and Turbidity Reduction group were very higher than model group(P<0.05).The expression and mRNA relative expression of the large and medium dose group and benzbromarone group were more higher(P<0.01).Conclusion:Through network pharmacology and basic research,the related mechanism of treating hyperuricemia was preliminarily revealed.Dehumidification and turbidification can act on hyperuricemia through many pathways,multiple targets and multiple signaling pathways,and it is found that the prescription of dehumidification and turbidification may be the purpose of treating hyperuricemia by upregulating the expression of renal ABCG2 protein.
