Objective Hyperuricemia in women with preeclampsia not only indicates a reminder of severity but also contributes directly to the pathogenesis of PE.ABCG2 Q141K and Q126X are closely associated with genetic susceptibility to hyperuricemia.Our aim was to investigate the genetic susceptibility of ABCG2 gene polymorphism to preeclampsia in parts of shangdong province han women.Methods A cohort of 793 preeclamptic han women(divided into 466 preeclampsia with hyperuricemia and 327 preeclampsia without hyperuricemia)and 744 normal pregnant han women were randomly recruited during January 2013 to June 2017.DNA was extracted to analyze the genotype and allele frequency of ABCG2 Q141K and Q126X by TaqMan real-time fluorescence quantitative PCR amplification.Statistical analysis was conducted by IBM SPSS Statistics 21.0 software.We conducted the differences of related basic data by one-way analysis of,of which statistically significant differences were further conducted by LSD t test.Hardy-Weinberg equilibrium was used to evaluate genotype of the control.Pearson chi-square test was used to detect the differences of allele and genotype distribution between the two groups(Fisher's test was used when the expected value was lower than 5).We used odds ratio and 95%confidence intervals to reveal relative risk degree.P<0.05(two-tailed)was considered to be statistically significant.Results The area under the curve of uric acid,urea nitrogen and creatinine was 0.886,0.776 and 0.745,respectively.The maximum point of UA Jorden index was318.75?mol/L.The age of the preeclampsia with hyperuricemia was lower than the preeclampsia without hyperuricemia and the control(P<0.001),while there was no statistical difference in age between the preeclampsia without hyperuricemia and the control(P>0.05).The uric acid level of the preeclampsia without hyperuricemia was higher than the control(P<0.001).Mean diastolic blood pressure in the preeclampsia was significantly higher than the control(P<0.001),while mean diastolic blood pressure in the preeclampsia with hyperuricemia was higher than the preeclampsia without hyperuricemia(P<0.001).The birth weight of preeclampsia was lower than the control,while that of preeclampsia with hyperuricemia was even lower than the preeclampsia without hyperuricemia(P<0.001).The frequency distribution of ABCG2 Q141K and Q126X in the control was in accordance with HWE(P>0.05).There was no significant difference between the preeclampsia and the control in the distribution of ABCG2 genotype(including dominant model and recessive model)and allele frequency(?~2=1.11,P=0.58 by genotype of Q141K;?~2=0.03,P=0.87 by dominant model of Q141K;?~2=1.09,P=0.30 by recessive model of Q141K;?~2=0.32,P=0.57 by allele of Q141K;P=0.33 by genotype of Q126X;P=0.33 by allele of Q126X).There was no statistically significant difference in the distribution of genotypes and allele frequencies at the ABCG2 Q141K and Q126X between the preeclampsia with hyperuricemia,the preeclampsia without hyperuricemia and the normal control group(Q141K:?~2=3.07,P=0.22 by genotype of preeclampsia with hyperuricemia;?~2=1.77,P=0.18 by allele of preeclampsia with hyperuricemia;?~2=0.47,P=0.79 by genotype of preeclampsia without hyperuricemia;?~2=0.42,P=0.52 by allele of preeclampsia without hyperuricemia;Q126X:P=0.72 by genotype of preeclampsia with hyperuricemia;P=0.72 by allele of preeclampsia with hyperuricemia;P=0.68 by genotype of preeclampsia without hyperuricemia;P=0.68 by allele of preeclampsia without hyperuricemia).There was no significant difference between mild/severe preeclampsia with hyperuricemia,mild/severe preeclampsia without hyperuricemia and normal pregnant women in the distribution of ABCG2 Q141K and Q126X genotype and allele frequency(P>0.05).There was also no significant difference between early-onset/late-onset preeclampsia with hyperuricemia,early-onset/late-onset preeclampsia without hyperuricemia and normal pregnant women in the distribution of ABCG2 Q141K and Q126X genotype and allele frequency(P>0.05).Conclusion There was no significant association between ABCG2 Q141K and Q126X polymorphisms and preeclampsia with hyperuricemia,mild/severe preeclampsia,early/late onset preeclampsia in parts of shandong province han women.Therefore,the polymorphisms of ABCG2 Q141K and Q126X may not be associated with preeclampsia in parts of shandong province han women.However,we should further explore their relationship with a larger scale.
