Objective:To summarize the clinical experience,identification ideas,academic thoughts and drug administration on the treatment of Oxaliplatin-induced peripheral neuropathy(OIPN)from Professor Jiege Huo.To investigate the potential mechanisms and therapeutic targets of Professor Huo’s core prescription,Huangqi Guizhi Wuwu Decoction(HQGZWWD),in the treatment of OIPN using network pharmacology and molecular docking techniques,in order to elucidate the effectiveness and scientific validity of Professor Huo’s core prescription.Methods:TCMSP,TargetNet database were used to screen and predict the effective compounds and targets of HQGZWWD.Genecard was used to identify the disease genes of OIPN;Venn diagrams were drawn to obtain the overlapping targets between HQGZWWD and OIPN,and network topology was performed using GeneMANIA to obtain potential therapeutic targets.Protein-protein interactions(PPI)netwok of OIPN genes were constructed by String platform,and Cytoscape was used to analyze and visualize the PPI netwok.Finally,the "HQGZWWD-effective compounds-overlapping targets-OIPN" network was constructed to explore the core targets of HQGZWWD for the treatment of OIPN and validated by molecular docking technique.DAVID was used for GO bioprocess and KEGG pathway enrichment analysis of targets.Results:A total of 48 herbal effective compounds,345 therapeutic targets and 28 OIPN disease genes were obtained after screening.Among them,29 therapeutic targets of HQGZWWD for OIPN treatment were obtained.PPI analysis identified core regulatory genes(TP53,EGFR,AKT1,IL6,MAPK3,CXCL8,etc.)of the herbal formulation in treating OIPN,and key effective compounds(quercetin,kaempferol,catechin,etc.)and core genes(PTGS2,ABCB1,ABCG2,CASP9,etc.)were found using Cytoscape.The Subsequent GO biofunctional and KEGG pathway enrichment analysis revealed that the pathological process of OIPN is related to inflammatory response and nerve injury,and HQGZWWD may play an interventional role in OIPN through multiple signaling pathways such as apoptosis,TNF,MAPK,and HIF-1.Conclusion:Our study confirmed that HQGZWWD has multi-compounds,multi-targets and multi-pathways in the treatment of OIPN through network pharmacology.In addition,HQGZWWD may be involved in anti-inflammation and promotion of nerve repair to improve OIPN,providing theoretical basis for the subsequent clinical and experimental studies of herbal formulation.
