Objective:1.To research the effects of Intradermal auricular electroacupuncture stimulation(iaES)with different frequencies on heart rate variability(HRV)of normal rats.2.To verify the efficacy of specific frequency iaES in the intervention of autonomic nervous imbalance model rats.Methods:一、Effects of different frequencies of iaES on HRV on normal rats1.Selection of stimulation site:left ear concha area of rats.2.Methods:A total of 40 male SD rats were randomly divided into 5 groups(n=8).The stimulation frequencies of 2Hz,15Hz,30Hz and 100Hz were selected and divided into four groups plus false electroacupuncture group.HRV was recorded before,during,and after the intervention.3.Detection of autonomic nerve function:the heart rate and lead electrocardiogram were recorded continuously during the experiment.Through software analysis,high frequency/low frequency(LF/HF),standardized low frequency(LFnu),standardized high frequency(HFnu),Total Power(TP),high frequency(HF),low frequency(LF),very low frequency(VLF)and heart rate(HR)were obtained.二、Effects of iaES at a specific frequency(30/100Hz)on autonomic nervous imbalance in rats.1.Establishment of the model of autonomic nervous imbalance:continuous intraperitoneal injections of clenbuterol(0.5mg/kg),acetylcholine(0.1mg/kg),propranolol(10mg/kg)and atropine(4mg/kg)were used to establish the sympathetic excitatory model,the parasympathetic excitatory model,the sympathetic inhibitory model and the parasympathetic inhibitory model.2.Experimental procedure:(1)HRV comparison of model rats and unmodel rats was conducted to verify the success of model building.(2)The effect of 30Hz and 100Hz iaES on autonomic nerve of model rats was verified by HRV comparison during and after intervention and before intervention,respectively.Results:一、Influence of iaES of different frequencies on HRV1.TP,HF and LF of iaES group increased in 20min after the intervention(P<0.05),HFnu increased in 20min after 2Hz iaES intervention(P<0.05),and TP decreased in 5min and 20min after 15Hz iaES intervention(P<0.05).There was no significant change in other indexes.2.30Hz iaES,VLF and HFnu in the intervention were higher than before,while LF/HF and LFnu were lower than before(P<0.05).After intervention,HF,VLF and TP continued to increase,while HR was lower than before intervention(P<0.05).3.100Hz iaES showed a decreasing trend in TP and HF 15min after intervention(P<0.05).In the intervention,HFnu was lower than before,while LF/HF and HR were significantly higher than before(P<0.05).二、Effect of 30Hz iaES on the model rats with autonomic nervous imbalanceEffects on sympathetic excitatory rats.After intervention,HF,LF and TP were significantly increased(P<0.05),while LF/HF and LFnu were significantly decreased(P<0.01,P<0.001).Effects on parasympathetic excitatory rats.LFnu and LF/HF increased both after and after intervention(P<0.05).HFnu decreased 25min after intervention(P<0.05)Effects on sympathetic inhibitory rats.VLF increased 30min after intervention(P<0.05).HF was lower than before at 10,15 and 20min after intervention,and TP was lower than before at 20min after intervention.LF/HF and LFnu increased significantly during and after intervention,while HFnu decreased significantly during and after intervention(P<0.05).Effect on parasympathetic nerve inhibition in rats.After 10min of intervention,HF and LF increased significantly(P<0.05).LFnu was significantly increased at 30min after intervention(P<0.05).HR decreased continuously from the intervention,and there was statistical difference 25 and 30min after intervention(P<0.05).三、Effects of 100Hz iaES on the model rats with autonomic nervous imbalanceEffects on sympathetic excitatory rats.TP,HF,LF,VLF and HFnu increased after intervention(P<0.05).HR decreased after intervention(P<0.05).Effects on parasympathetic excitatory rats.In the intervention,LF/HF and LFnu showed a significant upward trend,while HFnu showed a significant downward trend(P<0.05).After intervention,LF,VLF,HF and TP were significantly increased(P<0.05).Effects on sympathetic inhibitory rats.In the intervention,TP,HF,LF and LFnu were significantly increased,while HR was decreased at 20 and 25min(P<0.05).Effects on parasympathetic inhibitory rats.TP at 5min and 15min after intervention was higher than that before intervention,and VLF at 15min after intervention was higher than that before intervention(P<0.05).HFnu increased significantly during the intervention,while LF/HF decreased significantly and continued until after the intervention(P<0.05).LFnu was lower 15min after intervention than before.Conclusion:1.iaES influences HRV through state dependence and frequency dependence.30Hz and 100Hz iaES have their unique regulatory characteristics for different conditions of autonomic nervous imbalance.2.iaES with specific frequency has directivity in effect.iaES with different frequencies has different action directions on HRV.Among them,30Hz iaES can reduce heart rate by increasing vagal tone and affecting autonomic nerve.100Hz iaES can increase heart rate by enhancing sympathetic nerve activity and affecting autonomic nerve.3.The original state of the body plays a fundamental role in iaES driving HRV.100Hz iaES is more conducive to promoting autonomic balance in the condition of dominant excitability(sympathetic or parasympathetic).The inhibition of sympathetic or parasympathetic activity at 30Hz is more helpful to promote the balance of autonomic nerves.4.Whether it is 30Hz or 100Hz,iaES rebuilds autonomic balance by increasing the side of sympathetic/parasympathetic nerves that is relatively weak.The proportion of increase in sympathy and vagus is different,but the final direction(end point)is the balance of sympathy and vagus.
