| Objective: The study aims to explore whether there is a regular change of meridian hour in the levels of nitric oxide(NO),endothelin-1(ET-1)and B-type natriuretic peptide(BNP)from the perspective of meridian flow theory in comparison with healthy people and patients with chronic heart failure(CHF)due to coronary heart disease(CHD),and whether the variation of meridian hour in patients with CHF due to CHD is larger,in an attempt to provide a microcosmic mechanism reference for the causes of worsening of patients with CHF due to CHD at night,and to provide basis for timely intervention in the treatment of CHF due to CHD and improve the curative effect.Methods: Grouping: 20 patients with CHF due to CHD who met the diagnostic criteria of the Ruikang Hospital affiliated to Guangxi University of Traditional Chinese Medicine were selected as observation group.20 healthy people were selected as control group from the physical examination center of Ruikang Hospital affiliated to Guangxi University of Traditional Chinese Medicine.Specimen collection: The right elbow venous blood samples for 4ml each participants were collected at noon(11:00-13:00)and sub-time(23:00-01:00)on the same day,respectively,centrifuged for 10 min,the supernatants were extracted and stored from blood samples in the cryogenic refrigerator at-80℃.Specimen detection method: The level of NO in human serum was detected by Griess method while the levels of ET-1 and BNP in human serum were detected by ELISA.Results:(1)There was no statistically significant difference in age,sex and weight between the observation group and the control group,that is,the baseline was consistent,and there was comparability between the two groups.(2)Compared with the concentration of NO in the control group,the concentration of NO in the sub-time was lower than that at noon(P<0.05).The concentration of NO in the sub-time was also lower than that at noon in the observation group(P<0.05).The difference of meridian time in the both groups,that the observation group was smaller than that in control group(P <0.05).(3)Compared with the concentration of ET-1 in the control group,the concentration of ET-1 in the sub-time was higher than that at noon(P<0.05).The concentration of ET-1 in the sub-time was also higher than that at noon in the observation group(P<0.05).The difference of meridian time in the both groups,that the observation group was greater than that in control group(P <0.05).(4)Compared with the concentration of BNP in the control group,the concentration of BNP in the sub-time was higher than that at noon(P<0.05).The concentration of BNP in the sub-time was also higher than that at noon in the observation group(P<0.05).The difference of meridian time in the both groups,that the observation group was greater than that in control group(P <0.05).Conclusion: Regardless of the control group or the observation group,the concentrations of NO,ET-1 and BNP in the meridian time have changed horizontally,indicating that the levels of NO,ET-1,and BNP were different in both healthy people and patients with chronic heart failure of coronary heart disease,which is consistent with the time characteristics of Qi and blood’s prosperity and weakness of heart meridian;The concentration of NO in the sub-time in both groups was lower than that at noon,but the difference of meridian time in the observation group was lower than that in the control group,which indicated that the change of NO concentration in the observation group was weakened at meridian time;The concentration of ET-1 in the sub-time in both groups was higher than that at noon,but the difference of meridian time in the observation group was higher than that in the control group,which indicated that the change of ET-1 concentration in the observation group was increased at meridian time;The concentration of BNP in the sub-time in both groups was higher than that at noon,but the difference of meridian time in the observation group was higher than that in the control group,which indicated that the change of BNP concentration in the observation group was increased at meridian time;The the difference of meridian time of NO,ET-1 and BNP concentration is an important micro-mechanism in the pathogenesis of CHF due to CHD By intervening at night,increasing NO concentration,reducing ET-1 and BNP concentrations,and changing the circadian rhythm of the three to fine-tune them in a good direction may provide a new direction for the treatment of chronic heart failure of coronary heart disease. |
