| Background: Studies have shown that Hepatitis B X Protein(HBx)、Cytochrome P4502E1(CYP2E1)and β-catenin are associated with Hepatocellular carcinoma(HCC).However,it has not been reported that there are differences in the expression of HBx,CYP2E1 and β-catenin in HCC tissues with different degrees of differentiation,and there is no clear correlation among the three.Objective: To explore the correlation between the expression of HBx,CYP2E1 andβ-catenin in HBV-associated HCC and HCC,and to further clarify whether the three are related to the degree of malignancy of HCC.So as to provide a reference for further study on the pathogenesis of HCC.Method: 36 male hepatitis B related HCC specimens were collected from the Department of Pathology,Affiliated Hospital of Yan bian University from 2010 to 2020.HBx,CYP2E1 and β-catenin in HCC cells was observed by immunohistochemical staining method.Hcore scoring method was used to score the staining results of the three groups.According to the staining intensity of cytoplasm,no staining was 0 points,light yellow was 1 point,brown yellow was 2 points,and brown was 3 points.According to the number of positive cells : < 5% is 0 points,5%-25% is 1 point,26%-50% is 2 points,51%-75% is 3 points,>75% is 4 points.Multiply the two results:0 score is(-),1-4 score is(+),5-8 score is(+ +),9-12 score is(+ + +).The results showed that(-)was negative,"+" was weak positive,"+ +" was positive,and "+ +" was strong positive.Statistical analysis was used to study the correlation.Results:1.In 36 cases of HCC,there were 7 cases of highly differentiated HCC and 19 cases of poorly differentiated HCC2.The results of hcore scoring method: HBx was strongly positively expressed in 1case,positively expressed in 12 cases and weakly positively expressed in 4 cases in highly differentiated HCC;HBx was strongly positively expressed in 11 cases and positively expressed in 8 cases in poorly differentiated HCC.CYP2E1 was positively expressed in 2 cases,weakly positively expressed in 12 cases and negatively expressed in 3 cases in highly differentiated HCC;CYP2E1 was positively expressed in 8 cases,weakly positively expressed in 8 cases and negatively expressed in 3 cases in poorly differentiated HCC.β-catenin was expressed strongly positively in 1 case,positively in4 cases and weakly positively in 12 cases in highly differentiated HCC;β-Catenin was expressed strongly positively in 7 cases,positively in 11 cases and weakly positively in 1 case in poorly differentiated HCC.3.The chi-square test results: The strong positive expression rate of HBx in poorly differentiated HCC tissues was significantly increased,indicating that HBx was correlated with the degree of differentiation of HCC.The strongly positive expression rate of CYP2E1 was higher in poorly differentiated HCC tissues,and CYP2E1 was correlated with the differentiation degree of HCC.The strong positive expression rate of β-catenin in poorly differentiated HCC tissues was significantly increased,andβ-catenin was correlated with the differentiation degree of HCC.4.The spearman grade analysis results: there was no correlation between HBx andβ-catenin expression in highly differentiated HCC tissues(P > 0.05),HBx was positively correlated with β-catenin in poorly differentiated HCC tissues and all HCC tissues,and the expression trend was consistent(r=0.671,P < 0.05;r = 0.72,P <0.01).There was a positive correlation between the expression of CYP2E1 andβ-catenin in all HCC cells(r=0.577,P < 0.05;r= 0.863,P < 0.01;r= 0.704,P <0.01).HBx was positively correlated with CYP2E1 in highly differentiated,poorly differentiated and all HCC cells(r=0.814,P<0.01;r= 0.771,P<0.01;r = 0.756,P<0.01).Conclusion:1.The expression of HBx,CYP2E1 and β-catenin were correlated with the differentiation degree of HCC2.The expression of HBx and CYP2E1 were all positively correlated with β-catenin.The expression of HBx was positively correlated with that of CYP2E1.It is speculated that the three factors may play a synergistic role in the development of HCC. |
