eliminate cancer party PI3K/AKT/mTOR signaling pathway and the effect of PTEN,It provides a more direct and objective theoretical basis for the treatment of colorectal cancer and the inhibition of metastasis and proliferation of cancer cells,and provides a new perspective for the development of consensus on the diagnosis and treatment of colorectal cancer.Methods:1.The 20 pairs of sample tissues(20 cases of colorectal cancer and corresponding paraccancerous tissues respectively)provided by the Department of Pathology of Jiangsu Hospital of Traditional Chinese Medicine during 2018-2019 and 5 pairs of sample tissues(5 cases of colorectal cancer and corresponding paraccancerous tissues respectively)retained after colorectal cancer surgery in the anorectal department of our hospital during the five months from June 1 to October 31,2020 were used.Immunohistochemistry and q-PCR were used to detect the protein molecules of PTEN,PI3K,Akt and mTOR in each sample tissue and sample tissue,and the expression levels of each protein factor in colorectal cancer tissue and corresponding adjacent tissues were counted and the differences were calculated,which indirectly reflected whether each factor was involved in the development of human colorectal cancer.2.The subcutaneous tumor model of nude mice was established,the nude mice were killed,the tumor-bearing tissue was removed and weighed.The changes in tumor weight in each group were recorded and the differences were analyzed.Western Blot and immunohistochemical methods were used to detect the protein expression levels of PTEN,PI3K,Akt,mTOR,p-PI3K,p-Akt and p-mTOR in the samples of each group,and the differences among the groups were statistically analyzed.Results:1.In clinical trials,the results of immunohistochemistry showed that the expression of PTEN in paracancerous tissue was significantly higher than that in colorectal cancer group(P<0.05);the expression of PI3K,Akt and mTOR in paracancerous tissue was significantly lower than that in cancer tissue(P<0.05).The results of q-PCR showed that the protein contents of PTEN,PI3K,Akt and mTOR in colorectal cancer group were significantly lower than those in adjacent tissues(P<0.05).2.The results of tumor weight test in nude mice showed that:the average tumor weight of low-dose group was not significantly different from that of model group.(P>0.05);the average tumor weight of medium dose group and high-dose group of Guzheng Xiaoai formula were significantly lower than that of model group(P<0.05)3.In the animal experiment,Western blot results showed that:compared with the model group,the expression of PTEN in the low,medium and high dose groups of Guzheng Xiaoai Fang was significantly increased(P<0.05);the expression of p-PI3K/PI3K,p-Akt/Akt and p-mTOR/mTOR in the low,medium and high dose groups of Guzheng Xiaoai Fang was significantly decreased(P<0.05).The results of immunohistochemistry showed that:compared with the model group,the expression of p-pi3k,p-Akt and p-mTOR in the low-dose group of Guzheng Xiaoai decoction had a downward trend,but there was no statistical significance(P>0.05),The protein expressions of p-PI3K,p-Akt and p-mTOR in the middle and high dose groups of Guzheng Xiaoai prescription were significantly lower than those in the model group(P<0.05);compared with the model group,there was no significant difference in the expression of PTEN in the low and middle dose groups of Guzheng Xiaoai prescription(P>0.05),and the expression of PTEN in the high dose group was significantly higher than that in the model group(P<0.05);compared with the model group,the expression of PTEN in the low,middle and high dose groups of Guzheng Xiaoai prescription was significantly higher than that in the model group(P<0.05)The total expression levels of PI3K,Akt and mTOR in the three high-dose groups showed a downward trend,but there was no statistical significance(P>0.05)Conclusions:(1)The low expression of PTEN and the high expression of PI3K,Akt and mTOR in human colorectal cancer indicate that PI3K/Akt/mTOR signaling pathway is activated in the occurrence and development of human colorectal cancer,which is one of the key signal axes that we have been exploring to affect the pathogenesis of colorectal cancer.(2)Guzheng Xiaoai formula can inhibit the growth of implanted tumor in nude mice and the differentiation and proliferation of human colon cancer HT-29 cells.(3)Guzheng Xiaoai decoction can up regulate the expression of PTEN and reduce the expression of p-PI3K,p-Akt and p-mTOR in nude mice implanted tumor,and it is related to the concentration.High dose of Guzheng Xiaoai decoction has more effect.(4)guzheng Xiaoai
