Research background:Skin aging is the result of both internal and external factors,of which the main factor is photoaging caused by long-term exposure to ultraviolet radiation,which ultimately reduces the integrity of skin structure and destroys normal physiological functions,seriously affecting"beauty"and people’s healthy life.The adipose derived stem/stromal cells(ADSCs)is one of the source of adult progenitor cells.It is isolated from the adipose tissue,and it has the same multidirectional differentiation potential as bone marrow mesenchymal stem cells and peripheral blood stem cells.Because of the relatively abundant,easy separation,little injury to the body,strong self-renewal ability and low immunogenicity,The adipose derived stem/stromal cells(ADSCs)has become a very promising source of stem cells.Objective:To evaluate the efficacy and possibility of subcutaneous injection of ADSCs in the treatment of skin photoaging.Methods:1.To isolate and extracte ADSCs from adipose tissue of healthy adult women.2.Eighteen SPF grade female nude mice were randomly divided into control group(injected with PBS),UVB model group(injected with PBS 8 weeks after UVB irradiation),and ADSCs group(injected with PBS containing 1×10~6 ADSCs subcutaneously after 8 weeks of UVB irradiation),with 6 mice in each group.After feeding for 4 weeks,the back wrinkles of the nude mice were scored by taking photos to compare the changes in the gross appearance of the nude mice.The skin of the injection site on the back of the nude mice was cut and embedded into sections.Through HE staining observation of nude mouse skin structure and the thickness of dermis,through Masson staining to observe the arrangement and content of collagen fibers,the new blood vessels of skin were observed by CD31 immunohistochemistry,and the arrangement of elastic fibers was observed by resorcinol basic magenta staining.Results:1.ADSCs were successfully extracted from adipose tissue.ADSCs grow adherent and resemble fibroblasts.The results of oil red O,alizarin red and alkaline phosphatase staining were positive,indicating that the isolated ADSCs cells had the ability of adipogenic and osteogenic differentiation.Flow cytometry results showed high expression of CD29,CD44,CD90,CD105 and low expression of CD34,CD144,HLA-DR in ADSCs cells,which was consistent with the characteristics of ADSCs surface markers.2.Macroscopically,after 8 weeks of UVB irradiation,the naked mice showed obvious deep wrinkles on the back,with rough texture,thickened skin and even leather-like appearance,poor elasticity,With pigmentation and desquamation,etc.Compared with the normal control group,the wrinkle score was significantly increased.After injection of ADSCs for 4 weeks,the skin appearance of nude mice was significantly improved,with increased elasticity,soft texture,reduced wrinkles,no pigmentation,no desquamation,and the wrinkle score was significantly lower than that of UVB group.3.Histopathologically,compared with the UVB model group,after ADSCs injection,HE staining showed a significant increase in dermal thickness,Masson staining showed a significant increase in content of the collagen fibers,CD31immunohistochemical staining showed a significant increase in skin angiogenesis,and the resorcinol basic magenta staining showed an orderly arrangement of elastic fibers.Conclusion:The Adipose derived stem/stromal cells(ADSCs)can help restore the structural integrity of photoaged skin,promote skin collagen synthesis,make the elastic fiber rearrange,increase the thickness of the dermis and the number of new blood vessels.ADSCs can improve the signs of photoaging skin and ADSCs have a certain therapeutic effect on the photoaging skin of nude mice.
