Objective: To explore the correlation between rs1122608 of SMARCA4 gene and ischemic stroke,so as to provide reference for the prevention,diagnosis and treatment of ischemic stroke in Korean nationality in Yanbian area.Methods: 281 patients with IS in Yanbian Korean population were selected,including 159 males and 122 females,with an average age of 64.70 ± 9.94 years;At the same time,327 cases of age and gender matched non-stroke control group were randomly selected from the physical examination department of Yanbian Hospital,including 182 males and 145 females,with an average age of 63.42 ± 9.40 years old.After SNa Pshot technology for SNP typing detection,SPSS 25.0 for statistical analysis.Result:(1)Comparison of clinical parameters: There were no significant differences in age,gender,BMI,TC and HDL-C between the ischemic stroke group and the control group(P > 0.05).Diabetes,hypertension,TG and LDL-C in the ischemic stroke group were significantly higher than those in the control group,and the differences were statistically significant(P = 0.010,P = 0.002,P = 0.029,P = 0.049).(2)Hardy-Weinberg balance test: There was no significant difference between the allele frequency and genotype frequency of SMARCA4 gene rs1122608 in IS group and control group(P = 0.81,P = 0.19),in line with Hardy-Weinberg balance test,which shows that the two groups of samples selected in this study have good group representativeness.(3)Correlation analysis results between the IS group and the control group:(1)the frequency of T allele at rs1122608 of SMARCA4 gene in IS group was significantly lower than that in control group(P = 0.003),The G allele was used as a reference,suggesting that T allele was a protective factor with an OR value of 0.602(95%CI: 0.427~0.848).The genotype GG、GT and TT of rs1122608 locus of SMARCA4 gene in Korean population showed statistical differences between IS group and control group(P = 0.009),indicating that this locus is associated with ischemic stroke.(2)the results of genotype simulation showed that the TT+GT genotype in IS group was significantly lower than that in the control group(P = 0.002),and the OR value was0.557(95%CI: 0.383~0.810),suggesting that the TT+GT genotype in the Korean population in Yanbian area was a protective gene for ischemic stroke,presenting a dominant inheritance pattern.(4)Comparison of clinical parameters between different genotypes: The genotypes of the dominant model were compared with some clinical parameters,including BMI,TC,TG,HDL,LDL,diabetes,and hypertension,and there was no significant correlation between these clinical parameters and the genotypes of the dominant model(P > 0.05).(5)Analysis of IS-related risk factors: Since the dominance model(GT+TT)of rs1122608 locus of SMARCA4 gene in patients with ischemic stroke was significantly lower than that in the control group,only the dominance model was further analyzed here.The clinical parameters associated with ischemic stroke in Table 2,including diabetes,hypertension,TG,LDL-C and rs1122608 genotype GT+TT(GG genotype as reference),were taken as independent variables,and the dependent variable was defined as whether the patients had ischemic stroke(yes =1,no =0).Using the binary Logistic regression statistical analysis(P < 0.05),the statistics found that rs1122608GT+TT genotype P = 0.003,OR a value of 0.580(95% CI: 0.403~0.834),thus GT+TT genotype is a protective factor for the development of ischemic stroke in the Korean population in Yanbian area.Conclusion:(1)The G/T allele at rs1122608 of SMARCA4 gene is associated with the incidence of IS in the Korean population in Yanbian area,and the T allele is a protective factor at this locus,showing a dominant inheritance pattern.(2)The GT+TT genotypes at the rs1122608 locus of the SMARCA4 gene in Korean population in Yanbian area can reduce the risk of ischemic stroke and are protective factors for ischemic stroke.
