| Objective: In this study,the sera of subjects were collected to analyze the composition and content of phospholipid fatty acid profiles,and the levels of inflammatory cytokines IL-2,IL-6 and TNF-α in the serum were determined.Understanding the correlation between serum phospholipid unsaturated fatty acid profile,IL-2,IL-6,levels and coronary heart disease was investigated and the correlation of them in patients with coronary heart disease was further investigated.Methods:(1)Subjects: 330 patients(210 males 120 females)who were were admitted to the Department of Cardiology,the Second Affiliated Hospital of Dalian Medical University hospital from November 2019 to November 2020 due to chest tightness or chest pain were selected as subjects,aged from 40 to 80 years old,with an average age of(63.71±10.5)years old.On the basis of the results of coronary angiography,the subjects can be divided into CHD group and compared group.The patients with coronary heart disease who met the diagnostic criteria of coronary heart disease(≥1 coronary artery obstruction ≥50%)were the coronary heart disease group.The coronary angiography results showed no obvious stenosis or patients with coronary artery stenosis <50% are the compared group.Among them,there were 193 cases in the CHD group(138 males 55females)with an average age of(65.22±10.55)years old,and 137 cases(72 males 65females)in the compared group,with an average age of(61.58 ± 10.1)Years old.(2)Biochemical index examination: All the research subjects took venous blood on an empty stomach for at least 8 hours on the second morning.The specimens were collected and sent to the laboratory of our hospital for laboratory tests.Blood routine,troponin(CTNI),creatine kinase isoenzyme(CK-MB),renal function,hemagglutination routine,electrolyte,total cholesterol(TC),triglyceride(TG),LDL-C,high density lipoprotein cholesterol(HDL-C)and glycosylated hemoglobin were tested.(3)Fatty acid profile detection method: all the selected patients fasted for more than 8 hours in the next morning to take venous blood,and send the serum sample to the Institute of Chemical Physics,Chinese Academy of Sciences,and use gas chromatography analyzer to detect the fatty acid profile 31 fatty acid test data.(4)Detection of inflammatory factors: The detection of IL-2 and IL-6 by the determination of automatic electrochemiluminescence immunoassay;The detection of serum TNF-α is measured by enzyme-linked immunosorbent assay.The serum phospholipid unsaturated fatty acid profile,inflammatory factors IL-2,IL-6,and TNF-α levels of the coronary heart disease group and the control group were collected,and the data were analyzed by SPSS25.0 statistical software.Results:1.Compared with the control group,there is no statistical difference in general clinical data such as BMI,TG,HDL-C,LDL-C 、CNTI,etc.;Compared with the control group,there are more men and the age,the smoking history,hypertension,diabetes,TC,CK-MB,BNP,fasting blood glucose in the CHD group were significantly higher.2.Compared with the control group,the coronary heart disease group has the levels of total PUFA,total ω-3PUFA,EPA and DHA decreased significantly;and the EPA/AA ratio and DHA/AA ratio decreased significantly,while the ω6/ω3 ratio increased significantly.There was no statistical difference in SFA,MUFA and other PUFA in the serum phospholipid fatty acid profile.3.Compared with the compared group,the levels of IL-2,IL-6 and TNF-α in the CHD group were obviously higher than those in the compared group.4.Spearman correlation analysis was used to analyze the difference between fatty acid spectrum and Gensini score in CHD group and control group.The results showed that EPA level、total ω-3 PUFA level、EPA/AA ratio and DHA/AA ratio was negatively correlated with Gensini score,the correlation coefficients are r=-0.173 、r=-0.156、r=-0.184、r=-0.173(P<0.05)。And the ω6/ω3 ratio was positively correlated with Gensini score,the correlation coefficient is r=0.206(P<0.05);But There is no statistical significance between total PUFA 、DHA and Gensini points.5.The results of Spearman correlation analysis of IL-2,IL-6,TNF-α and Gensini scores showed that the level of IL-2,IL-6,TNF-αwas positively correlated with the Gensini score of coronary artery disease,and the correlation coefficient are r=0.245、r=0.190、r=0.209,and the difference was statistically significant(P<0.01).6.Spearman correlation analysis was used to analyze the difference between fatty acid spectrum and IL-2,IL-6,TNF-α in CHD group and control group.The results showed that EPA level was negatively correlated with IL-2 and IL-6;while the ratio of ω6/ω3 was positively correlated with IL-2 and IL-6 levels;and the ratio of EPA/AA was negatively correlated with the level of IL-2.While there is no correlation between fatty acid index and TNF-α。Conclusions:1.In the serum phospholipid unsaturated fatty acid profile,the total PUFA level,total ω-3PUFA level,EPA level,DHA level and EPA/AA ratio,DHA/AA ratio are negatively correlated with CHD;while the ω6/ω3 ratio is positively correlated with CHD.2.Serum levels of ω-3PUFA,EPA levels,EPA/AA ratio and DHA/AA ratio in patients with CHD are negatively correlated with the severity of CHD,while ω6/ω3 ratio was positively correlated with CHD.3.The levels of IL-2,IL-6 and TNF-α were correlated with CHD and positively correlated with the severity of CHD.4.The serum EPA level in the coronary heart disease group was negatively correlated with IL-2 and IL-6 levels;the ratio of ω6/ω3 in CHD was Positively related to IL-2 and IL-6 levels;the ratio of EPA/AA in CHD Positively related to IL-2 levels. |
