| Objective:In this experiment,we observed the effect of Baoyinjian on the function damage of human chorionic trophoblast HTR-8/Svneo cell line induced by phospholipid antibody,and studied the proliferation,invasion and apoptosis of damaged cells,to preliminarily explore the possibility of Baoyinjian improving the function of damaged trophoblast in early pregnancy,and to discuss its scientific theoretical basis,so as to provide the basis for the production of optimizing the treatment of traditional Chinese medicine and clinical application of Baoyinjian in the treatment of antiphospholipid antibody positive recurrent abortion.Methods:1.HTR-8/Svneo cells were cultured in vitro.The cells were divided into control group and experimental group.The experimental group was cultured with different concentrations of Baoyinjian(0,200,400,600,800μg/ml)for 24 hours.The activity of HTR-8/Svneo cells was detected by CCK-8.2.The water extract of Baoyinjian(400μg/ml)was prepared and HTR-8/Svneo was cultured in vitro.The effect of water extract of Baoyinjian on proliferation inhibition of HTR-8/Svneo cells induced by phospholipid antibody was detected by CCK-8.3.The water extract of Baoyinjian(400μg/ml)was prepared and HTR-8/Svneo was cultured in vitro.The inhibition of cell invasion was detected by Transwell chamber invasion test.4.The water extract of Baoyinjian(400μg/ml)was prepared and HTR-8/Svneo was cultured in vitro.The effect of Baoyinjian on trophoblast apoptosis induced by antiphospholipid antibody was detected by flow cytometry.Results:1.The results of CCK-8 test showed that Baoyinjian at the final concentration of 200μg/ml to 800μg/ml could promote the proliferation of HTR-8/Svneo cells,and the effect of 400μg/ml group was the most significant(P<0.01);Baoyinjian at the final concentration of 800μg/ml had cytotoxicity(P<0.01),and inhibited cell proliferation.2.The CCK-8 experiment results show that antiphospholipid antibody inhibited the proliferation of HTR-8/Svneo cells(P<0.01),and Baoyinjian could promote the proliferation of damaged HTR-8/Svneo cells(P<0.01).With the increase of culture time(24h,48h),the inhibition of antibody on the proliferation of HTR-8/Svneo cells increased gradually,and the improvement of Baoyinjian on the proliferation of damaged HTR-8/Svneo cells was more obvious.3.The Transwell invasion test results show that antiphospholipid antibody could inhibit the invasion of HTR-8/Svneo cells(P<0.01),and Baoyinjian could improve the invasion inhibition of damaged cells(P=0.007<0.01).4.The flow cytometry results show that antiphospholipid antibody increased the apoptosis of HTR-8/Svneo cells(P<0.01),and Baoyinjian could improve the apoptosis of damaged cells(P<0.01).Conclusion:Antiphospholipid antibody can increase trophoblast proliferation,invasion and apoptosis,and Baoyinjian can improve the proliferation,inhibition of invasion function and apoptosis of trophoblast caused by antiphospholipid antibody. |
